山羊CMKLR1基因的克

【摘要】:試驗旨在克隆山羊CMKLR1基因序列并進行生物信息學分析,研究其表達與肌內(nèi)脂肪(IMF)沉積的相關(guān)性。采集山羊皮下脂肪組織,應用RT-PCR克隆山羊CMKLR1基因序列;以GAPDH(GenBank登錄號:AJ431207.1)為內(nèi)參基因,實時熒光定量PCR檢測其在山羊不同組織中的表達量及在肌肉組織中的時序表達譜(以肝臟表達量為基準);測定肌肉組織中的IMF含量,分析其與CMKLR1mRNA表達量的相關(guān)性。結(jié)果顯示,克隆獲得了山羊CMKLR1基因,1 089bp的CDS區(qū),GenBank登錄號:KT165374。實時熒光定量PCR分析顯示,CMKLR1基因在24月齡山羊心臟、肝臟、脾臟、肺臟、腎臟、脂肪、背最長肌、股二頭肌和臂三頭肌中均存在不同程度的表達,其中在肺臟中表達量顯著高于其他組織(P0.05);CMKLR1基因在1~3和24月齡的山羊背最長肌、臂三頭肌和股二頭肌中表達趨勢相同,均是在股二頭肌中表達量最高,而8~10月齡則是在臂三頭肌中表達量最高。山羊IMF含量以24月齡背最長肌含量最高。山羊背最長肌、股二頭肌和臂三頭肌中CMKLR1mRNA表達與IMF含量相關(guān)性不顯著(P0.05)。CMKLR1基因不參與山羊IMF沉積作用,試驗結(jié)果為進一步研究CMKLR1基因奠定理論基礎(chǔ)。
[Abstract]:The aim of the experiment was to clone goat CMKLR1 gene sequence and analyze its expression by bioinformatics, and to study the correlation between the expression of goat CMKLR1 gene and intramuscular fat (IMF) deposition. The goat subcutaneous adipose tissue was collected and goat CMKLR1 gene sequence was cloned by RT-PCR. Using GAPDH(GenBank accession number AJ431207.1 as internal reference gene, real-time fluorescence quantitative PCR was used to detect its expression in different tissues of goat and its temporal expression profile in muscle tissue. The content of IMF in muscle tissue was measured. The results showed that the CMKLR1 gene of goat was cloned and cloned into GenBank accession number: 1: KT165374.The real-time fluorescence quantitative PCR analysis showed that the CMKLR1 gene was expressed in heart, liver, spleen, lung, kidney, fat of goat at 24 months of age. The expression of CMKLR1 gene in the longissimus dorsi muscle, biceps femoris muscle and triceps arm muscle was significantly higher than that in other tissues, and the expression trend of CMKLR1 gene in the triceps arm and biceps femoris muscle was the same. The highest expression was found in biceps femoris muscle, and the highest expression was found in triceps arm muscle at the age of 8 to 10 months. The IMF content of goat was the highest in the 24 month old longissimus dorsi muscle, and the highest in the goat dorsal longissimus muscle at the age of 8 to 10 months. There was no significant correlation between CMKLR1mRNA expression and IMF content in biceps femoris and triceps arm muscle. The results of the experiment laid a theoretical foundation for further study of CMKLR1 gene.
