【摘要】：本研究旨在建立一種可同時(shí)檢測(cè)8種常見牛傳染病病原體(口蹄疫病毒、藍(lán)舌病病毒、水皰性口炎病毒、牛病毒性腹瀉病毒、牛輪狀病毒、產(chǎn)腸毒大腸桿菌、牛傳染性鼻氣管炎病毒和小反芻獸疫病毒)的GeXP高通量快速檢測(cè)方法。將多重PCR技術(shù)和毛細(xì)管電泳技術(shù)有效地結(jié)合,根據(jù)上述8種病原體的基因保守序列,設(shè)計(jì)并合成了8對(duì)特異性引物,在每對(duì)特異性引物的5′端加上一段通用引物,形成特異性嵌合引物。優(yōu)化反應(yīng)體系和反應(yīng)條件,用單一病毒和混合病毒樣品來驗(yàn)證所建立的GeXP方法的特異性,用克隆質(zhì)粒稀釋液來測(cè)試GeXP方法的靈敏度,檢測(cè)305份臨床樣品,進(jìn)一步驗(yàn)證該方法的可靠性。結(jié)果:優(yōu)化后的GeXP檢測(cè)體系,可擴(kuò)增出各病原體對(duì)應(yīng)的特異片段,能在100拷貝·μL~(-1)水平同時(shí)并特異地檢測(cè)出8種牛病病原體核酸。305份臨床樣品的檢測(cè)結(jié)果與熒光PCR檢測(cè)結(jié)果一致,陽(yáng)性樣品測(cè)序結(jié)果表明無(wú)非特異性擴(kuò)增的假陽(yáng)性。結(jié)果顯示,所建立的GeXP方法具有高通量、特異性好、靈敏度高、方便快速等優(yōu)點(diǎn),可用于臨床鑒別診斷和流行病學(xué)調(diào)查,具有較高的臨床應(yīng)用價(jià)值。
[Abstract]:The purpose of this study was to establish a novel pathogen for simultaneous detection of 8 common bovine infectious diseases (foot-and-mouth disease virus, bluetongue virus, vesicular stomatitis virus, bovine viral diarrhea virus, bovine rotavirus, enterotoxigenic Escherichia coli). High throughput GeXP rapid detection method for bovine infectious rhinotracheitis virus and small ruminant ruminant virus. The multiplex PCR technique and capillary electrophoresis technique were effectively combined, according to the gene conserved sequence of the above eight pathogens, Eight pairs of specific primers were designed and synthesized. A segment of universal primers was added to the 5 'end of each pair of specific primers to form a specific chimeric primer. The reaction system and reaction conditions were optimized. Single virus and mixed virus samples were used to verify the specificity of the established GeXP method. The sensitivity of the GeXP method was tested with clone plasmid dilution solution, and 305 clinical samples were detected. Results: the optimized GeXP detection system can amplify the specific fragments corresponding to each pathogen. At the level of 100 copies 路渭 L ~ (-1), nucleic acid of 8 kinds of bovine disease pathogens, nucleic acid, was detected at the same time and the results were consistent with the results of fluorescent PCR. The results of sequencing of positive samples showed that there was no nonspecific amplification of false positive, and the results showed that, The established GeXP method has the advantages of high throughput, good specificity, high sensitivity, convenience, rapidity, etc. It can be used in clinical differential diagnosis and epidemiological investigation, and has high clinical application value.
