高谷物日糧促進(jìn)山羊瘤胃上皮單羧酸轉(zhuǎn)運(yùn)蛋白1及鈉鉀ATP酶mRNA的表達(dá)

【摘要】:本試驗(yàn)旨在研究高谷物日糧對(duì)山羊瘤胃上皮形態(tài)結(jié)構(gòu)及單羧酸轉(zhuǎn)運(yùn)蛋白(monocarboxylate transporter,MCT)和鈉鉀ATP酶mRNA表達(dá)的影響。將10頭裝有永久性瘤胃瘺管的健康閹割公山羊隨機(jī)分為飼喂全粗料日糧的對(duì)照組(Hay,0%谷物,n=5)和飼喂高谷物日糧的處理組(HG,65%谷物,n=5),試驗(yàn)期為7周。試驗(yàn)開始后,于每周晨飼后的0、2、3、4、6、8和12h連續(xù)采集瘤胃液監(jiān)測(cè)瘤胃pH值的變化,收集其中第0、3、6和12h的瘤胃液待測(cè)揮發(fā)性脂肪酸(volatile fatty acid,VFA)濃度。試驗(yàn)的第50天,屠宰采集瘤胃上皮用于形態(tài)學(xué)及基因定量分析。研究結(jié)果顯示:與全粗料組山羊相比,高谷物組山羊瘤胃pH值、乙酸濃度及乙丙比都顯著下降(P0.001),而瘤胃丙酸濃度、丁酸濃度及其他VFA濃度都顯著升高(P0.001);高谷物日糧組的瘤胃乳頭長度顯著高于對(duì)照組(P=0.001),瘤胃乳頭寬度顯著低于對(duì)照組(P0.001),但是兩組間的瘤胃乳頭表面積并無顯著差異;透射電鏡結(jié)果顯示,長期飼喂高谷物日糧導(dǎo)致瘤胃上皮細(xì)胞線粒體發(fā)生降解;實(shí)時(shí)定量PCR結(jié)果表明,與對(duì)照組相比,高谷物日糧顯著升高了MCT1(P0.001)和鈉鉀ATP酶(P=0.001)的mRNA表達(dá)量,顯著降低了MCT4的mRNA表達(dá)量(P=0.041),但對(duì)MCT2的表達(dá)沒有顯著影響(P=0.305);進(jìn)一步分析這些基因的mRNA表達(dá)量與pH值和VFA濃度之間的相關(guān)性,結(jié)果顯示,MCT1和鈉鉀ATP酶的mRNA表達(dá)量與瘤胃pH值和乙酸濃度呈顯著負(fù)相關(guān),與總VFA、丙酸、丁酸的含量呈顯著正相關(guān),而MCT4的mRNA表達(dá)量與pH值呈顯著正相關(guān),與總VFA、丙酸、丁酸的含量呈顯著負(fù)相關(guān)。以上結(jié)果提示:高精料引起的瘤胃pH值下降和VFA的變化可能與瘤胃上皮MCT和鈉鉀ATP酶表達(dá)量的變化相關(guān)。研究結(jié)果對(duì)深入認(rèn)識(shí)高谷物飼喂引發(fā)的瘤胃功能紊亂具有重要意義。
[Abstract]:The purpose of this study was to study the effects of high grain diet on the morphological structure of rumen epithelium and the expression of monocarboxylate transporterase mRNA and monocarboxylate transporterase mRNA in goats. Ten healthy castrated male goats with permanent rumen fistula were randomly divided into feeding. The control group (Hayan 0%) and the treatment group fed high grain diet (HGN 65%) were used for 7 weeks, the trial period was 7 weeks. The rumen fluid was collected continuously for monitoring the rumen pH value after feeding every morning, and the concentration of volatile volatile fatty acid (volatile fatty acid) volatile fatty acido (VFA) in rumen fluid was collected at the 0th and 12h after feeding continuously. The concentration of volatile fatty acid volatile fatty acid (volatile fatty acid) in the rumen fluid was determined on the 50th day of the experiment, and the concentration of volatile fatty acid (VFA) in the rumen fluid was determined at 12 h and 0 h after feeding. The results showed that the rumen pH value, acetic acid concentration and ethylene-propyl ratio in the high grain group decreased significantly compared with the whole crude diet group, while the rumen propionic acid concentration decreased significantly in the slaughtering and collecting rumen epithelium, while the rumen propionic acid concentration in the high grain group was significantly lower than that in the whole crude diet group. Butyric acid concentration and other VFA concentrations were significantly higher than that of control group (P 0.001), the rumen papilla length of high grain diet group was significantly higher than that of control group, and the rumen papilla width was significantly lower than that of control group, but there was no significant difference in rumen papilla surface area between the two groups. The results of transmission electron microscope showed that the degradation of mitochondria in rumen epithelial cells was induced by long-term feeding of high grain diet, the results of real-time quantitative PCR showed that the high grain diet significantly increased the mRNA expression of MCT1 + P0.001) and sodium and potassium ATP enzyme P0. 001) compared with the control group. The mRNA expression of MCT4 was significantly decreased, but the expression of MCT2 was not significantly affected. The correlation between the mRNA expression of these genes and pH value and VFA concentration was further analyzed. The results showed that the mRNA expression of MCT1 and sodium and potassium ATP was negatively correlated with rumen pH and acetic acid concentration, and positively correlated with the contents of total VFA, propionic acid and butyric acid, while the mRNA expression of MCT4 was positively correlated with pH value, and with total VFA-propionic acid. The results suggested that the decrease of rumen pH and the change of VFA might be related to the changes of MCT and ATP expression in rumen epithelium. Rumen dysfunction caused by feeding is of great significance.
