【摘要】：顆粒細(xì)胞凋亡與卵泡閉鎖是顆粒細(xì)胞功能基因及凋亡相關(guān)基因的表達(dá)結(jié)果,通過手術(shù)法獲取三種不同直徑的牛卵泡(小型卵泡2 mm;中型卵泡2~6 mm;大型卵泡6mm),分離卵泡顆粒細(xì)胞并提取總RNA,以研究不同直徑卵泡中顆粒細(xì)胞相關(guān)基因的表達(dá)。結(jié)果表明,牛卵巢上小型卵泡顆粒細(xì)胞中Fshr的表達(dá)顯著低于中型和大型卵泡(P0.05),而中型和大型兩者之間差異不顯著(P0.05);中型卵泡和小型卵泡顆粒細(xì)胞中CYP19的相對表達(dá)顯著低于大型卵泡(P0.05);中型卵泡和小型卵泡顆粒細(xì)胞中促細(xì)胞凋亡基因BAX的表達(dá)顯著低于大型卵泡(P0.05),而在中型和大型卵泡顆粒細(xì)胞中抑制細(xì)胞凋亡基因BCL2的表達(dá)顯著低于小型卵泡(P0.05),且隨著卵泡的增大而下調(diào);Casapase 8與Caspase3的表達(dá)模式相同,其表達(dá)量均隨著卵泡直徑的增加顯著上升(P0.05)。隨著卵泡生長與直徑增加,卵泡內(nèi)部分顆粒細(xì)胞雖然在繼續(xù)增殖,部分顆粒細(xì)胞已經(jīng)開始凋亡,卵泡逐漸進(jìn)入閉鎖階段。
[Abstract]:Granulosa cell apoptosis and follicular atresia are the results of expression of granulosa cell functional genes and apoptosis-related genes. Three kinds of bovine follicles with different diameters (small follicles 2 mm; medium follicles 2 mm; large follicles 6 mm) were obtained by surgical method. Granulosa cells were isolated from follicular granulosa cells and total RNAs were extracted to study the expression of genes related to granulosa cells in follicles with different diameters. The expression of Fshr in small follicle granulosa cells on bovine ovary was significantly lower than that in medium and large follicular granulosa cells, but there was no significant difference between medium and large follicles, and the relative expression of CYP19 in medium and small follicular granulosa cells was significantly lower than that in large follicular granulosa cells. The expression of apoptosis-promoting gene BAX in medium and small follicular granulosa cells was significantly lower than that in large follicular granulosa cells, while the expression of BCL2 in medium and large follicular granulosa cells was significantly lower than that in small follicular granulosa cells. The expression pattern of Casapase 8 and Caspase3 was the same with the increase of follicle. With the growth and diameter of follicle, some granulosa cells continued to proliferate, some granulosa cells began to apoptosis and follicle gradually entered atresia stage.
