GnIH對(duì)母鵝繁殖性能和公雞生殖發(fā)育的影響

【摘要】:Gn IH可以作用于垂體前葉或作用于Gn RH來(lái)抑制促性腺激素的合成與釋放,是調(diào)節(jié)動(dòng)物繁殖性能的重要生殖激素之一。目前用生殖免疫調(diào)控的方法提高家畜繁殖力已受到廣泛關(guān)注。許多研究應(yīng)用抑制素、催乳素、Gn IH等生殖激素作用于動(dòng)物體內(nèi)或體外實(shí)驗(yàn)均可發(fā)現(xiàn)其參與調(diào)控動(dòng)物的生長(zhǎng)繁殖。因此,本實(shí)驗(yàn)主要以馬崗鵝和公雞為研究對(duì)象,從外源性給予Gn IH和主動(dòng)免疫Gn IH兩種不同的研究方法進(jìn)行研究:實(shí)驗(yàn)一選取健康的成年母鵝36只,平均分成3組。第一組僅在d1給予30%PVP-K30溶解的Gn IH溶液,給予Gn IH 1 mg/只;第二組在d1、d4、d7給予生理鹽水溶解的Gn IH溶液,給予Gn IH 1 mg/只;第三組在d1、d4、d7給予生理鹽水,給予1 m L每只。試驗(yàn)中記錄母鵝每日產(chǎn)蛋數(shù)和就巢情況;并在d1、d4、d7、d10定時(shí)采集血清,在試驗(yàn)結(jié)束時(shí)屠宰母鵝,采集下丘腦和垂體組織樣和檢查卵泡發(fā)育情況。研究表明給予產(chǎn)蛋母鵝外源性Gn IH能一定程度上抑制自身Gn IH的基因表達(dá),降低血液中LH濃度,提高雌二醇濃度,主要通過(guò)抑制中小卵泡發(fā)育,進(jìn)而在一定程度上抑制母鵝產(chǎn)蛋。實(shí)驗(yàn)二選取30只處于就巢期健康的成年馬崗鵝母鵝,平均分成免疫組和對(duì)照組(n=15),試驗(yàn)在正式開(kāi)始時(shí),對(duì)母鵝進(jìn)行人工強(qiáng)制終止就巢。免疫組分別在d1、d21、d41接種含Gn IH蛋白的疫苗1 m L/只(以此含1.0、1.0、0.5 mg的Gn IH蛋白),對(duì)照組在d1、d21、d41注射未含免疫原的生理鹽水制備的乳化苗。兩組鵝試驗(yàn)開(kāi)始飼養(yǎng)于短光照(11L:13D)下以繼續(xù)維持正常產(chǎn)蛋,在d72將每天的光照時(shí)間延長(zhǎng)至18小時(shí)(18L:6D)至d95試驗(yàn)結(jié)束。試驗(yàn)期記錄鵝的就巢發(fā)生、每日產(chǎn)蛋數(shù),并每20天對(duì)母鵝采血一次,分離血清留待激素測(cè)定。在試驗(yàn)結(jié)束時(shí)對(duì)兩組母鵝進(jìn)行屠宰,檢查卵泡發(fā)育情況,并采集下丘腦和垂體樣本,留待基因表達(dá)水平檢測(cè)。研究結(jié)果表明對(duì)成年母鵝主動(dòng)免疫Gn IH可誘導(dǎo)體內(nèi)產(chǎn)生特異性抗Gn IH的抗體,抑制Gn IH的基因表達(dá),略微提高了Gn RH、VIP和PRL的基因表達(dá),調(diào)節(jié)血清中LH、E2、P4等相關(guān)生殖激素進(jìn)而提高母鵝的產(chǎn)蛋性能。另外,延長(zhǎng)光照會(huì)抑制母鵝的產(chǎn)蛋,但其對(duì)對(duì)照組的作用比免疫組更強(qiáng),這可能是因?yàn)橹鲃?dòng)免疫Gn IH對(duì)母鵝的產(chǎn)蛋促進(jìn)作用還在維持。實(shí)驗(yàn)三選120只60日齡健康公雞,隨機(jī)平均分成兩組,免疫組和對(duì)照組,所有雞均采用籠養(yǎng),每籠2只。免疫組接種含Gn IH蛋白的疫苗1 m L/只(含0.5 mg的Gn IH蛋白),對(duì)照組注射未含免疫原的生理鹽水制備的乳化苗。分別在試驗(yàn)的d1、d21和d41(即60、80和100日齡)進(jìn)行胸部肌肉注射接種疫苗。在試驗(yàn)期,從60日齡開(kāi)始,每隔20天進(jìn)行對(duì)公雞進(jìn)行采血,所得血清保存于-20℃,留待測(cè)定血液中抗體水平和激素水平;每隔30天分別從兩組隨機(jī)挑選8只公雞進(jìn)行屠宰,采取睪丸組織并檢測(cè)其發(fā)育情況,并采集下丘腦和垂體留待基因表達(dá)水平檢測(cè)。從195日齡開(kāi)始兩組公雞進(jìn)行人工采精訓(xùn)練(按摩法),待一周建立條件反射后,每隔兩天采精一次進(jìn)行精液品質(zhì)檢測(cè)。研究結(jié)果表明,對(duì)發(fā)育期公雞主動(dòng)免疫Gn IH能有效的刺激了機(jī)體產(chǎn)生特異性抗Gn IH抗體,并在整個(gè)實(shí)驗(yàn)過(guò)程中維持較高水平。免疫Gn IH后能抑制Gn IH、VIP、PRL的基因表達(dá),提高LH的基因表達(dá),進(jìn)而促進(jìn)睪丸發(fā)育和血液循環(huán)中睪酮、INH、ACT的分泌,提高精液品質(zhì)。
[Abstract]:Gn IH can function in the anterior pituitary function in Gn or RH to inhibit the synthesis and release of gonadotropin, is one of the most important reproductive hormone regulation of animal reproductive performance. The current method for reproductive immune regulation to improve the fecundity of cattle has attracted widespread attention. Many studies by prolactin inhibin, Gn, IH and other reproductive hormones in animal in vivo or in vitro experiments can be found which is involved in the regulation of animal growth and reproduction.
