【摘要】：策勒黑羊具有全年發(fā)情、多胎的特點(diǎn),是新疆策勒縣特有的優(yōu)良綿羊品種。為研究與保護(hù)策勒黑羊遺傳資源,為后續(xù)開(kāi)展逆轉(zhuǎn)錄酶聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)、斑點(diǎn)雜交試驗(yàn)和構(gòu)建c DNA文庫(kù)試驗(yàn)等提供條件,試驗(yàn)采用經(jīng)典異硫氰酸胍(Trizol)法提取策勒黑羊卵巢總RNA,通過(guò)核酸蛋白儀測(cè)定所提取RNA在不同波長(zhǎng)下的吸光度值,計(jì)算RNA的提取量與純度,并利用瓊脂糖凝膠電泳顯示提取結(jié)果和驗(yàn)證RNA的完整性。結(jié)果表明:采用Trizol法提取得到的策勒黑羊卵巢RNA的濃度、純度與完整性均可達(dá)到后續(xù)試驗(yàn)的要求,可為后續(xù)策勒黑羊種質(zhì)資源保護(hù)和利用等生物學(xué)試驗(yàn)的開(kāi)展提供參考依據(jù)。
[Abstract]:Cele Black Sheep has the characteristics of full year estrus and multiple births, and is a unique fine sheep breed in Cele County, Xinjiang. In order to study and protect the genetic resources of Cele Black Sheep, To provide conditions for reverse transcriptase polymerase chain reaction reverse transcriptase polymerase chain reaction reverse transcriptase polymerase chain reaction reverse transcriptase polymerase chain reaction RT-PCRN, dot blot hybridization and construction of c DNA library. In this experiment, the classical guanidine thiocyanate (Trizol) method was used to extract the total RNA from the ovary of the black sheep. The absorbance of the extracted RNA at different wavelengths was determined by nucleic acid protein analyzer, and the extraction amount and purity of RNA were calculated. Agarose gel electrophoresis was used to show the extraction results and verify the integrity of RNA. The results showed that the concentration, purity and integrity of ovarian RNA extracted by Trizol method could meet the requirements of subsequent tests. It can provide reference for the following biological experiments such as conservation and utilization of Cele black sheep germplasm resources.
