菖蒲龍骨散對雛雞免疫應(yīng)激的預(yù)防作用及機(jī)理研究

【摘要】:本試驗(yàn)旨在研究菖蒲龍骨散對LPS致雛雞免疫應(yīng)激的預(yù)防作用及機(jī)理,篩選出最佳組方為臨床防治雛雞免疫應(yīng)激提供理論依據(jù)。將1日齡健康雛雞180只,隨機(jī)平均分成6組,每組30只(分3個(gè)重復(fù),每個(gè)重復(fù)10只)。其中,A組為空白對照組,B組為LPS組,C組為LPS+芪參散組,D組為LPS+白術(shù)茯苓散組,E組為LPS+菖蒲龍骨散組,F組為LPS+氯丙嗪組。7日齡時(shí),A、B組飼喂基礎(chǔ)日糧,C、D、E和F組飼喂添加各組組方的基礎(chǔ)日糧,飼喂一星期,在14、16、18、20日齡時(shí),對A組雛雞腹腔注射生理鹽水,每只1 ml;對B、C、D、E和F組雛雞腹腔注射LPS+生理鹽水,每只1 ml,LPS用量為1600μg/kg(BW)。各組分別于14、21、28、35日齡時(shí)稱重,心臟采取EDTA抗凝血,測定紅、白細(xì)胞總數(shù),心臟采血離心得血清,測定血液指標(biāo),隨后剖殺采集胸腺、脾臟、法氏囊,測定其重量,計(jì)算免疫器官指數(shù),取法氏囊制作石蠟切片進(jìn)行HE染色,觀察并攝像。試驗(yàn)結(jié)果表明,(1)白術(shù)茯苓散、菖蒲龍骨散和氯丙嗪能夠有效預(yù)防免疫應(yīng)激引起的雛雞生產(chǎn)性能下降,菖蒲龍骨散的效果更好。(2)白術(shù)茯苓散、菖蒲龍骨散和氯丙嗪對免疫應(yīng)激引起的雛雞血清總蛋白和白蛋白降低都有一定的預(yù)防作用,但相比來說添加菖蒲龍骨散的效果更好。(3)菖蒲龍骨散對免疫應(yīng)激引起的雛雞血清IL-1、IL-6升高的預(yù)防作用最好。(4)白術(shù)茯苓散和菖蒲龍骨散對免疫應(yīng)激引起的雛雞血清Ig A、Ig M、Ig G的升高都有預(yù)防作用。(5)對免疫應(yīng)激引起的雛雞血清皮質(zhì)酮升高,白術(shù)茯苓散和菖蒲龍骨散都有較好的預(yù)防效果。結(jié)論,菖蒲龍骨散對雛雞免疫應(yīng)激的預(yù)防作用最好。這為菖蒲龍骨散作為飼料添加劑在養(yǎng)雞業(yè)中的應(yīng)用提供了理論依據(jù)。
[Abstract]:The purpose of this study was to study the preventive effect and mechanism of Acorus Longgu Powder on immune stress induced by LPS in chicks, and to select the best formula to provide theoretical basis for clinical prevention and treatment of immune stress in chicks. 180 healthy chicks of 1 day old were randomly divided into 6 groups. 30 rats in each group (3 repeats, Among them, group A is a blank control group, group B is LPS group, group C is LPS Qi Shen San group, group D is LPS Atractylodes Fuling powder group, group E is LPS Acorus Longgu powder group, group F is LPS chlorpromazine group, group B is fed basic diet at day 7. Group E and F were fed with basal diet supplemented with each group. The chickens in group A were given intraperitoneal injection of normal saline (1 ml / ml) for one week, and the chickens of group A and group F were injected intraperitoneally with LPS. The dosage of LPS was 1 600 渭 g / kg BWN. Each group was weighed at 1421 835 days, and the heart was treated with EDTA anticoagulant. The total number of red and white blood cells were measured, the serum was collected from the heart, and the blood indexes were measured. Then, the thymus, spleen and bursa of Fabricius were collected, their weight was measured, the immune organ index was calculated, and paraffin sections of the bursa of Fabricius were taken to make paraffin sections for HE staining. The experimental results showed that Atractylovir Fuling Powder, Acorus Longgu Powder and chlorpromazine could effectively prevent the performance decline of chicks caused by immune stress, and the effect of Acorus Longgu Powder was better than that of Atractylodes Fuling Powder. Acorus Longgu Powder and chlorpromazine have some preventive effects on the decrease of serum total protein and albumin induced by immune stress. But the effect of Acorus Longgu Powder was better than that of Acorus Longgu Powder.) Acorus Longgu Powder had the best preventive effect on the increase of IL-1 and IL-6 in the serum of chicks induced by immune stress. The best one was Baizhu Fuling Powder and Acorus Longgu Powder on the serum Ig of chicks induced by immune stress. The increase of Ig mg Ig G had a preventive effect on serum corticosterone level in chicks induced by immune stress. Both Atractylodes Atractylodes Fuling Powder and Acorus Longgu Powder have good preventive effects. Conclusion Acorus Longgu Powder has the best preventive effect on immune stress in chicks, which provides a theoretical basis for the application of Acorus Longgu Powder as feed additive in chicken industry.
