FABP3及FABP4基因SNP檢測及其與秦川肉牛肉質(zhì)性狀的關(guān)聯(lián)性分析

【摘要】:脂肪酸結(jié)合蛋白3(fat acid binding proteins 3,FABP3)和脂肪酸結(jié)合蛋白4(fat acid binding proteins 4,FABP4)是FABPs家族的重要成員。FABPs的主要作用是調(diào)節(jié)脂肪酸攝取和細(xì)胞內(nèi)轉(zhuǎn)運(yùn),盡管蛋白質(zhì)序列有很大的差異,但基因結(jié)構(gòu)相似。本研究旨在探索FABP3及FABP4基因?qū)η卮ㄅH庥眯缕废?以下簡稱“秦川肉牛”)肉質(zhì)性狀的影響,為秦川牛肉用選育提供理論基礎(chǔ)。為了研究FABP3及FABP4基因?qū)η卮ㄈ馀H赓|(zhì)性狀的影響,本試驗(yàn)首先采用生物信息學(xué)方法分析了FABP3及FABP4基因的生物學(xué)特征及生物進(jìn)化信息。然后選取了飼養(yǎng)條件接近的571頭18~36月齡的秦川肉牛作為試驗(yàn)材料,進(jìn)行PCR擴(kuò)增,采用直接測序法,運(yùn)用DNAstar軟件對測序結(jié)果進(jìn)行對比分析,尋找突變位點(diǎn);并與秦川肉牛肉質(zhì)性狀(背膘厚、眼肌面積和肌內(nèi)脂肪含量)關(guān)聯(lián)分析,以便為秦川肉牛選育改良工作篩選合適的分子標(biāo)記,選擇出具有優(yōu)良性狀的組合基因型,為秦川肉牛的科學(xué)選育提供理論支持。本研究的主要結(jié)果如下:1.FABP3基因序列編碼133個(gè)氨基酸;FABP4基因序列編碼132個(gè)氨基酸。生物信息學(xué)分析表明:該家族的基因序列在哺乳動物不同物種中具有較高的相似性。2.FABP3基因第3內(nèi)含子上存在1個(gè)SNP位點(diǎn)g.60298CT;FABP4基因第1內(nèi)含子上存在2個(gè)SNP位點(diǎn)g.2686AT和g.2834CG,第4外顯子上存在1個(gè)SNP位點(diǎn)g.4220AG。只有g(shù).2686AT存在兩種基因型,分別是AA和AT基因型,TT基因型缺失。3.FABP3基因g.60298CT位點(diǎn)TT基因型個(gè)體均值顯著高于CT和CC基因型(P0.05)。FABP4基因g.2834CG位點(diǎn)CC基因型肉質(zhì)性狀表現(xiàn)最優(yōu),眼肌面積均值比GG基因型均值高7.541cm2,差異極顯著(P0.01);而肌內(nèi)脂肪含量均值比GG基因型均值高0.254%,差異顯著(P0.05)。4.FABP4基因g.4220AG位點(diǎn)TT基因型為優(yōu)良基因型,在背膘厚方面,TT基因型顯著高于CC基因型(P0.05)。5.FABP4基因3個(gè)SNP位點(diǎn)的優(yōu)勢單倍型組合為H1H1(ACA-ACA),其個(gè)體的眼肌面積極顯著高于單倍型組合H3H3(P0.01);個(gè)體肌內(nèi)脂肪含量均值極顯著高于單倍型組合H3H7(P0.01)。因此,在本試驗(yàn)群體中,單倍型組合H1H1可被視為一種最為優(yōu)秀的單倍型組合。綜上,FABP3及FABP4基因與秦川肉牛肉質(zhì)性狀緊密關(guān)聯(lián),可作為影響肉質(zhì)性狀的有效候選基因用于標(biāo)記輔助選擇,有助于秦川肉牛定向高效選育和群體遺傳改良。
[Abstract]:Fatty acid binding protein 3fat acid binding proteins 3 (FABP3) and fatty acid binding protein 4fat acid binding proteins 4 (FABP4) are important members of FABPs family. FABPs regulate fatty acid uptake and intracellular transport, although protein sequences vary greatly. The purpose of this study was to explore the effects of FABP3 and FABP4 genes on the meat quality traits of a new line of Qinchuan Beef (Qinchuan Beef cattle). In order to study the effects of FABP3 and FABP4 genes on meat quality of Qinchuan beef cattle, The biological characteristics and evolution information of FABP3 and FABP4 genes were analyzed by bioinformatics, and then 571 Qinchuan beef cattle (1836 months old) with similar feeding conditions were selected for PCR amplification. Direct sequencing method was used to compare and analyze the results of sequencing using DNAstar software to find mutation sites, and to analyze the correlation with meat quality traits (backfat thickness, eye muscle area and intramuscular fat content) of Qinchuan beef cattle. In order to select suitable molecular markers for the breeding and improvement of Qinchuan Beef cattle, and select combination genotypes with excellent traits, The main results of this study are as follows: 1. FABP3 gene sequence encodes 133 amino acids and FABP4 gene encodes 132 amino acids. Bioinformatics analysis shows that the gene sequence of this family is as follows: 1. 2. There is a SNP locus in intron 3 of FABP3 gene. 2. There are two SNP loci (g.2686AT and g.2834CG) on intron 1 of CTP4 gene of FABP3 gene, and one SNP locus (g.4220AGG) on exon 4 of FABP3 gene. Only g.2686AT in intron 1 of FABP3 gene. There are two genotypes, 3. The individual mean values of TT genotype at CT locus of FABP3 gene were significantly higher than those of CT genotypes and CC genotypes (P0.05N. FABP4 gene, g.2834CG), respectively. The mean area of eye muscle was 7.541 cm 2 higher than that of GG genotype, and the difference was very significant (P 0.01), while the mean value of intramuscular fat content was 0.254% higher than that of GG genotype. The difference was significant (P 0.05) .4.TT genotype at g.4220 AG of FABP4 gene was a good genotype. In terms of backfat thickness, the dominant haplotype combinations of the three SNP loci of the FABP4 gene were significantly higher than those of the CC genotype (P 0.05), and the area of the individual eye muscle was significantly higher than that of the haplotype combination H _ 3H _ 3P _ (0.01), and the mean value of intramuscular fat content was significantly higher than that of the haplotype combination H _ 3H _ 3H _ 3P _ (0.01). Haplotype combination H3H7, P0.01. therefore, In this population, haplotype H1 can be regarded as the best haplotype combination. FABP3 and FABP4 genes are closely related to the meat quality traits of Qinchuan Beef cattle, and can be used as effective candidate genes for marker-assisted selection of meat quality traits in Qinchuan Beef cattle. It is helpful to direct and efficient breeding and population genetic improvement of Qinchuan beef cattle.
