【摘要】：為了探究黃藤素的體外細(xì)胞毒性,采用不同濃度的黃藤素作用山羊子宮內(nèi)膜上皮細(xì)胞(EECs)后,采用MTT法繪制細(xì)胞生長(zhǎng)曲線;倒置顯微鏡下觀察細(xì)胞生長(zhǎng)狀態(tài);瑞氏—姬姆薩染色法觀察細(xì)胞形態(tài)學(xué)變化并通過(guò)測(cè)定乳酸脫氫酶(LDH)含量檢測(cè)細(xì)胞膜的完整性。結(jié)果顯示,當(dāng)黃藤素的濃度為10~227μg/mL時(shí),黃藤素對(duì)EECs有促增殖的作用;當(dāng)濃度大于227μg/mL時(shí)出現(xiàn)細(xì)胞增殖抑制現(xiàn)象,藥物對(duì)細(xì)胞的半數(shù)增殖抑制率(IC50)為360μg/mL;瑞氏—姬姆薩染色結(jié)果顯示,100、200μg/mL的黃藤素作用于EECs時(shí)均未對(duì)其細(xì)胞核、細(xì)胞漿的形態(tài)產(chǎn)生影響;LDH結(jié)果顯示,藥物濃度≥250μg/mL時(shí)細(xì)胞LDH的釋放量極顯著高于正常細(xì)胞組(P0.01)。綜上所述,黃藤素作用于山羊EECs后呈現(xiàn)一定的毒性作用,且呈現(xiàn)顯著的劑量依賴(lài)效應(yīng),作用于EECs的安全劑量范圍為≤250μg/mL。
[Abstract]:In order to investigate the cytotoxicity of sinomenin in vitro, the growth curve of goat endometrial epithelial cells (EECs) was drawn by MTT method, and the growth state of the cells was observed under inverted microscope. The morphological changes of the cells were observed by Ricker-Giemsa staining and the integrity of the cell membrane was determined by measuring the content of lactate dehydrogenase. The results showed that when the concentration of sinomenin was 10 ~ 227 渭 g / mL, it could promote the proliferation of EECs.
