GLP實驗動物設(shè)施屏障環(huán)境消毒方法的研究

【摘要】:本研究選擇了山東省食品藥品檢驗所藥物安全評價中心在日常消毒工作中經(jīng)常使用的84消毒液、苯扎溴銨、百毒殺三種消毒劑作為研究對象。三者為含氯類、含季銨鹽類具有代表性的消毒液。通過不同濃度的消毒劑在不同作用時間的條件下,使用浸泡、擦拭、噴霧三種消毒措施測定消毒效果。通過試驗了解三種消毒劑對GLP實驗動物設(shè)施屏障環(huán)境與飼養(yǎng)用具物品的消毒效果。力求尋找理想的消毒劑和與它匹配的消毒方式。得到試驗結(jié)果:三種消毒劑在浸泡消毒方法中,均適用于GLP實驗動物設(shè)施屏障環(huán)境中飼養(yǎng)器具(飼養(yǎng)鼠盒、飲水瓶)浸泡消毒。浸泡消毒30min都能達到99.9%以上的滅菌率。有效氯含量為2000mg/l的84消毒液,濃度為0.1%苯扎溴銨,稀釋比例為1:100的百毒殺浸泡消毒效果比較好。三種消毒劑浸泡消毒效果與濃度、浸泡作用時間均成正比例關(guān)系。三種消毒劑在噴霧消毒方法中,試驗結(jié)果證明84消毒液、苯扎溴銨、百毒殺均對屏障環(huán)境中墻面與環(huán)境空氣噴霧消毒有一定的消毒和抑菌的效果,其中有效氯含量1000mg/l的84消毒液,濃度為0.05%的苯扎溴銨,稀釋比例1:600的百毒殺噴霧消毒效果比較好。三種消毒劑在擦拭消毒方法中,試驗結(jié)果說明如果每天對GLP屏障環(huán)境中墻面與籠架臺面擦拭消毒一次,使用有效率含量500mg/l、1000mg/l、2000mg/l的84消毒液;濃度為0.05%、0.1%的苯扎溴銨;稀釋比例為1:600、1:200、1:100的百毒殺均可以達到99.9%以上的滅菌率;如果每兩天對GLP屏障環(huán)境中墻面與籠具表面擦拭一次。使用有效氯含量1000mg/l、2000mg/l的84消毒液;濃度為0.1%苯扎溴銨;稀釋比例為1:600、1:200、1:100的百毒殺可以達到99.9%以上滅菌率。采用稀釋比例為1:100的百毒殺可以維持99.9%的滅菌率5天。試驗中有效氯含量為1000mg/l的84消毒液、濃度為0.1%苯扎溴銨以及稀釋比例為1:100的百毒殺擦拭消毒效果最好。上述結(jié)果證明三種消毒劑都比較適用GLP實驗動物設(shè)施屏障環(huán)境中消毒工作。浸泡消毒,滅菌效果好的為有效氯濃度2000mg/l的84消毒液、濃度為0.1%苯扎溴銨、稀釋比例為1:100的百毒殺。因84消毒液具有腐蝕性,籠具中的籠蓋等不銹鋼制品消毒建議使用苯扎溴銨與百毒殺浸泡消毒。但從三種消毒劑浸泡消毒效果對比中,84消毒液與百毒殺優(yōu)于苯扎溴銨消毒劑,更適合浸泡消毒方式。噴霧消毒如選擇每日一次時,可選用有效氯含量500mg/l的84消毒液、濃度為0.1%苯扎溴銨、稀釋比例為1:600百毒殺可以達到99.9%消毒效果。擦拭消毒三者效果均很可靠,其中百毒殺效果最好。如果每日擦拭一次可選用有效氯含量500mg/l~2000mg/l的84消毒液、濃度為0.05%~0.01%苯扎溴銨、稀釋比例為1:600以上的百毒殺。兩天擦拭消毒一次,可選用有效氯含量1000mg/l~2000mg/l的84消毒液、濃度為0.1%苯扎溴銨,稀釋比例1:600以上的百毒殺。五天擦拭消毒一次可選用1:100的百毒殺。根據(jù)比較不同消毒方式下三種消毒劑的消毒試驗結(jié)果,綜合辯證的考慮三種消毒液的安全性、實用性以及性價比等條件。得出這三種消毒劑均比較適用于GLP實驗動物設(shè)施消毒工作。實際消毒工作中,常使用噴霧消毒與擦拭消毒相結(jié)合的方式,其消毒效果比較好。
[Abstract]:This study selected 84 disinfectant safety evaluation center of Shandong province food and drug inspection of drugs often used in daily disinfection work, benzalkonium bromide, three kinds of disinfectant Baidusha as the research object. Three for chlorine, disinfection solution containing quaternary ammonium salt representative. By varying the concentration of the disinfectant in different effect of time under the conditions of use, soak, wipe, spray three kinds of disinfection measures determined the disinfection effect. Understand the three kinds of disinfectants on GLP experimental animal facility barrier environment and feeding utensils disinfection effect by experiment. In order to find the ideal disinfectant and its matching disinfection. Test results have been obtained: three kinds of disinfectants in immersion disinfection in the method, are applicable to the environmental GLP experimental animal facility barrier rearing apparatus (feeding rat box, water bottle) immersion disinfection. Immersion disinfection 30min can achieve sterilization rate of more than 99.9%. Effect of chlorine content of 84 disinfectant solution 2000mg/l, concentration of 0.1% benzalkonium bromide, the dilution ratio of 1:100 Baidusha immersion disinfection effect is better. Three kinds of disinfection disinfectant soaking and concentration, soaking time effect is proportional. The results show that three kinds of disinfectants are more suitable environment GLP Experimental Animal Barrier Facilities and disinfection work. But from the comparison of three kinds of disinfection disinfectant, disinfectant Baidusha is better than that with 84 benzalkonium bromide disinfectant, more suitable for immersion disinfection. Such as the choice of daily disinfection spray once, 84 disinfectant can choose effective chlorine content of 500mg/l, the concentration of 0.1% benzalkonium bromide, the dilution ratio of 1:600 Baidusha can achieve the disinfection effect of 99.9%. Three disinfection effects are very reliable, which Baidusha best. If a daily wipe 84 disinfectant can choose effective chlorine content of 500mg/l~2000mg/l the concentration of 0.05%~0.01% of benzalkonium bromide, the dilution ratio is more than 1:600 Baidusha. Two days cleaning disinfection, 84 disinfectant can choose effective chlorine content of 1000mg/l~2000mg/l, the concentration of 0.1% benzalkonium bromide, dilution ratio With more than 1:600 Baidusha. Five days a 1:100 can be used to wipe disinfection Baidusha. According to the test results of the three kinds of disinfectant disinfection under different disinfection methods, comprehensive dialectic to consider the safety of three kinds of disinfectant, practical and cost-effective conditions. The three kinds of disinfectant were suitable for sterilization GLP experimental animal facility. The actual work of disinfection, disinfection and disinfection spray is often used in combination, the disinfection effect is good.
