【摘要】：[目的]探討泰妙菌素生產(chǎn)菌種原生質(zhì)體的最優(yōu)制備方法。[方法]通過對原生質(zhì)體制備條件進(jìn)行優(yōu)化,建立泰妙生產(chǎn)菌種原生質(zhì)體制備體系。[結(jié)果]優(yōu)化后制備條件:以培養(yǎng)3 d的菌絲體為材料,將其置于含質(zhì)量濃度0.10%溶壁酶、0.60 mol/L MgSO_4的酶液中,25℃酶解3.0~3.5 h,將酶解液用G2砂芯漏斗過濾,濾液即為原生質(zhì)體懸液。采用該方法,泰妙生產(chǎn)菌種原生質(zhì)體制備率達(dá)1.5×10~6個/m L。[結(jié)論]試驗結(jié)果為提高泰妙菌素生產(chǎn)菌種的產(chǎn)量提供了理論依據(jù)。
