【摘要】:[目的]探討泰妙菌素生產(chǎn)菌種原生質體的最優(yōu)制備方法。[方法]通過對原生質體制備條件進行優(yōu)化,建立泰妙生產(chǎn)菌種原生質體制備體系。[結果]優(yōu)化后制備條件:以培養(yǎng)3 d的菌絲體為材料,將其置于含質量濃度0.10%溶壁酶、0.60 mol/L MgSO_4的酶液中,25℃酶解3.0~3.5 h,將酶解液用G2砂芯漏斗過濾,濾液即為原生質體懸液。采用該方法,泰妙生產(chǎn)菌種原生質體制備率達1.5×10~6個/m L。[結論]試驗結果為提高泰妙菌素生產(chǎn)菌種的產(chǎn)量提供了理論依據(jù)。
