【摘要】:為研究胰島素樣生長(zhǎng)因子IGF-I及其受體IGF-IR在雞胚胎腿部肌肉發(fā)育過(guò)程中的作用,選取藏雞和矮小蛋雞受精蛋孵化后,采集雞胚胎孵化第9天、第13天和第20天時(shí)腿部肌肉組織,并利用熒光定量PCR技術(shù)檢測(cè)IGF-I和IGF-IR mRNA的表達(dá)水平,同時(shí)檢測(cè)IGF-I基因編碼區(qū)的單核苷酸多態(tài)性并利用RFLP(限制性片段長(zhǎng)度多態(tài)性)方法對(duì)篩選到的單核苷酸多態(tài)性進(jìn)行群體水平分型。結(jié)果表明:藏雞和矮小蛋雞胚胎腿部肌肉組織在孵化第9天和第13天時(shí)的IGF-I和IGF-IR的mRNA表達(dá)量均極顯著高于第20天,且藏雞IGF-I的mRNA表達(dá)量在第9天也顯著高于第13天,而矮小蛋雞的IGF-I mRNA表達(dá)水平在第9天和第13天無(wú)明顯差異。此外,在整個(gè)孵化過(guò)程中,矮小蛋雞的IGF-IR的表達(dá)模式與IGF-I表達(dá)模式基本一致。而藏雞IGF-IR的表達(dá)水平在第9天明顯高于第13天但未達(dá)到顯著水平(P0.05),同時(shí)發(fā)現(xiàn)第9天藏雞IGF-IR的表達(dá)水平也高于矮小蛋雞(P0.05)。除此之外,在藏雞和矮小蛋雞的IGF-I的編碼區(qū)均發(fā)現(xiàn)c.282AG單核苷酸多態(tài)性,該SNP在藏雞中主要以雜合子AG基因型為優(yōu)勢(shì)基因型,基因型頻率為0.57,而在矮小蛋雞中主要以GG基因型為優(yōu)勢(shì)基因型,基因型頻率為0.77。根據(jù)試驗(yàn)結(jié)果分析得出,IGF-I和IGF-IR在腿部肌肉組織的表達(dá)水平在雞胚胎孵化中期(第9天和第13天)顯著高于孵化后期(第20天),可能是由于中期屬于胚胎腿部肌肉快速發(fā)育階段而后期腿部肌肉發(fā)育變得相對(duì)遲緩,同時(shí)IGF-I和IGF-IR的表達(dá)水平也隨之發(fā)生變化。
[Abstract]:In order to study the role of insulin-like growth factor (IGF-I) and its receptor IGF-IR in the muscle development of the legs of chicken embryos, Tibetan and dwarf laying eggs were selected and hatched on the 9th day after hatching. The expression levels of IGF-I and IGF-IR mRNA were detected by fluorescence quantitative PCR on the 13th and 20th day. Single nucleotide polymorphisms in the coding region of IGF-I gene were also detected and RFLP (restriction fragment length polymorphism) was used. Methods the selected single nucleotide polymorphisms were classified at population level. The mRNA expression of IGF-I and IGF-IR in the leg muscle of Tibetan chicken and dwarf laying hens on the 9th and 13th day of hatching was significantly higher than that on the 20th day. The mRNA expression of IGF-I in Tibetan chickens was also significantly higher on the 9th day than on the 13th day, but there was no significant difference in the IGF-I mRNA expression level of the dwarf layers on the 9th and 13th days. In addition, there was no significant difference in the expression of IGF-I mRNA between the two groups on the 9th day and the 13th day. Throughout the incubation process. The expression pattern of IGF-IR in dwarf layers was basically consistent with that of IGF-I, while the expression of IGF-IR in Tibetan chickens was significantly higher than that in day 13 on the 9th day (P < 0.05). P0.05). At the same time, it was found that the expression level of IGF-IR in Tibetan chickens on the 9th day was also higher than that in dwarf layers. The single nucleotide polymorphism of c.282 AG was found in the coding region of IGF-I of Tibetan and dwarf layers, and the dominant genotype of SNP was heterozygous AG genotype in Tibetan chickens. The genotype frequency was 0.57, while the GG genotype was the dominant genotype and the genotype frequency was 0.77 in the dwarf laying hens. The expression levels of IGF-I and IGF-IR in the muscle tissue of the legs were significantly higher than those in the late hatching period (day 9 and day 13). This may be due to the rapid development of the leg muscles in the middle stage of the embryo and the relatively slow development of the leg muscles in the later stages. Meanwhile, the expression levels of IGF-I and IGF-IR also change.
