【摘要】:綿羊脂肪沉積的部位和沉積方式顯著影響羊肉品質(zhì)和養(yǎng)羊效益。因此,揭示功能基因、日糧及生長(zhǎng)階段對(duì)綿羊脂肪沉積的影響,對(duì)改良綿羊肉品質(zhì)具有重要意義。研究選取了脂肪代謝相關(guān)功能基因NELL1和FMO3,比較其在不同尾型綿羊品種(哈薩克羊、灘羊、同羊、西藏羊)尾部脂肪組織中的表達(dá)差異;探討營養(yǎng)水平及生長(zhǎng)階段對(duì)其在灘羊脂肪組織中表達(dá)的影響,旨在揭示不同營養(yǎng)水平及不同生長(zhǎng)階段脂尾型綿羊NELL1和FMO3基因表達(dá)的組織特異性及其表達(dá)模式,為脂尾型綿羊體內(nèi)脂肪沉積的營養(yǎng)調(diào)控提供理論依據(jù)。試驗(yàn)一:不同品種綿羊尾脂中NELL1和FMO3 mRNA表達(dá)差異分析選擇健康、體重相近的9月齡灘羊、同羊、哈薩克羊和西藏羊羯羊各3只,采集尾部脂肪組織,利用實(shí)時(shí)熒光定量PCR技術(shù),檢測(cè)NELL1和FMO3 mRNA表達(dá),結(jié)果如下:1.FMO3mRNA在哈薩克羊尾脂中表達(dá)量最高(P0.05);在同羊、灘羊、西藏羊尾脂中依次下降,且差異顯著(P0.05)。2.NELL1mRNA在西藏羊尾脂中表達(dá)量最高(P0.05);在灘羊、同羊、哈薩克羊中表達(dá)量依次下降。試驗(yàn)二:營養(yǎng)水平對(duì)不同階段灘羊部分組織中NELL1和FMO3 mRNA表達(dá)的影響選擇健康4月齡灘羊112只隨機(jī)分為4組,公母各半,每組四個(gè)重復(fù),每個(gè)重復(fù)7只羊。試驗(yàn)根據(jù)增重目標(biāo)分為3個(gè)階段:第一階段(22~28kg),第二階段(28~35kg),第三階段(35~40kg),每個(gè)階段各組分別飼喂不同營養(yǎng)水平的日糧,由低到高分為T1、T2、T3、T4組。每個(gè)生長(zhǎng)階段結(jié)束時(shí)采集肝臟、腎周脂肪、尾脂、皮下脂肪的組織樣品,提取樣品總RNA,采用實(shí)時(shí)熒光定量PCR的方法分析NELL1和FMO3 mRNA的表達(dá)差異。結(jié)果如下:1.NELL1 mRNA在尾脂中的表達(dá),第1階段以T4組為最高(P0.05),其他三組差異不顯著;第2階段和第3階段,也都是以T4組表達(dá)量最高(P0.05),T1、T2、T3組表達(dá)量略有波動(dòng)。2.NELL1 mRNA在皮下脂中的表達(dá)。第1階段和第2階段四個(gè)營養(yǎng)水平組間表達(dá)差異不顯著(P0.05);第3階段,T4組表達(dá)量最高(P0.05),依次為T2、T3、T1組。相同營養(yǎng)水平下,NELL1 mRNA在第1階段表達(dá)量最高(P0.05)。3.NELL1 mRNA在腎周脂中的表達(dá)。在同一生長(zhǎng)階段,營養(yǎng)水平對(duì)灘羊腎周脂肪組織中NELL1mRNA的表達(dá)無顯著影響(P0.05);在相同營養(yǎng)水平下,不同生長(zhǎng)階段間表現(xiàn)出表達(dá)差異顯著(P0.05)。4.NELLL1 mRNA在肝臟中的表達(dá)。第1階段,在四個(gè)營養(yǎng)水平組間表達(dá)差異不顯著;第2階段,表達(dá)以T4組最高(P0.05),依次為T3、T1、T2組;第3階段,表達(dá)以T1組為最高(P0.05),依次為T2、T3、T4組。3個(gè)階段表達(dá)依次升高,其中第3階段表達(dá)量最高(P0.05)。5.FMO3 mRNA的表達(dá)。尾部脂肪組織中最高表達(dá)均出現(xiàn)在T2組;在第1階段、第2階段和第3階段,T1、T2、T3、T4組中表達(dá)量均呈現(xiàn)先升高再降低的趨勢(shì);皮下脂肪組織T3組最高,腎周脂最高表達(dá)出現(xiàn)在T1組,肝臟的最高表達(dá)出現(xiàn)在第T4組,最高表達(dá)組與相同階段時(shí)的最低表達(dá)組相比差異顯著的(P0.05)。試驗(yàn)三:NELL1、FMO3mRNA在灘羊不同組織中的表達(dá)差異分析在試驗(yàn)第三階段結(jié)束當(dāng)天,選取T2組4只灘羊進(jìn)行屠宰,采集肝臟、腎周脂肪、尾脂、皮下脂肪組織,利用實(shí)時(shí)熒光定量PCR技術(shù),檢測(cè)NELL1、FMO3mRNA的相對(duì)表達(dá)量,結(jié)果如下:1.NELL1 mRNA在灘羊尾部脂肪組織、皮下脂肪組織、腎周脂肪組織、肝臟中均有表達(dá)。NELL1在肝臟組織中表達(dá)最高,顯著高于尾脂、皮下脂和腎周脂(P0.05),其次是在腎周脂中的表達(dá),高于尾脂和皮下脂(P0.05),在尾脂和皮下脂中的表達(dá)最低,兩者之間差異不顯(P0.05)。2.FMO3 mRNA在灘羊尾部脂肪組織、皮下脂肪組織、腎周脂肪組織、肝臟中均有表達(dá)。FMO3在肝臟組織中表達(dá)最高,顯著高于尾部脂肪組織、皮下脂肪組織和腎周脂肪組織(P0.05),其次是在尾部脂肪組織中的表達(dá),高于腎周脂肪組織和皮下脂肪組織(P0.05),在腎周脂肪組織和皮下脂肪組織中的表達(dá)最低,兩者之間差異不顯(P0.05)。綜合以上試驗(yàn)結(jié)果,表明NELL1和FMO3基因在綿羊組織中的表達(dá)受到品種、部位、生長(zhǎng)階段、營養(yǎng)水平等原因會(huì)有不同的表達(dá)結(jié)果,其表達(dá)差異是與脂肪的沉積差異是存在一定關(guān)系。
[Abstract]:Location and mode of deposition of sheep fat deposition significantly affect the quality of mutton and sheep benefits. Therefore, to reveal the functional gene, the effects of dietary and growth stages of sheep fat deposition, and has the important significance for improving the quality of sheep meat. The fat metabolism related genes NELL1 and FMO3, the tail type in different sheep breeds (Kazak sheep, Tan sheep, Tong sheep, Tibet sheep) differentially expressed in adipose tissue of the tail; nutrition level and growth effects on its expression in sheep adipose tissue, to reveal the different nutritional levels and different growth stages of fat tail sheep NELL1 and expression of FMO3 gene and tissue specific expression pattern.
