【摘要】：為研究卵巢靜止奶牛與發(fā)情奶牛體內(nèi)差異代謝物變化,選取產(chǎn)后60~90d的年齡、胎次、體況相近的健康經(jīng)產(chǎn)高產(chǎn)奶牛作為實(shí)驗(yàn)動物。根據(jù)奶牛發(fā)情表現(xiàn)、直腸檢查和B超檢查及激素檢測的結(jié)果,選取10頭奶牛為發(fā)情組(A),10頭奶牛為卵巢靜止組(B),然后對兩組奶牛血漿樣品進(jìn)行無偏性的1 H譜核磁共振檢測,對比其血漿代謝組學(xué)圖譜,進(jìn)行多元統(tǒng)計(jì)分析,篩選及鑒定卵巢靜止奶牛血漿差異代謝產(chǎn)物,并進(jìn)行生物信息學(xué)分析。結(jié)果顯示:相對于正常發(fā)情奶牛,卵巢靜止奶牛血漿中共12種代謝產(chǎn)物表現(xiàn)異常,包括水平增加的乙酸、檸檬酸和酪氨酸,以及水平下降的低密度脂蛋白、極低密度脂蛋白、脂質(zhì)、丙氨酸、丙酮酸、肌酸、膽堿、磷酸膽堿和甘油磷酸膽堿等;這些代謝產(chǎn)物與糖代謝、氨基酸代謝、脂蛋白代謝和膽堿代謝的異常密切相關(guān);通過參與生物合成通路調(diào)節(jié),干擾了奶牛正常卵泡生長。并且發(fā)現(xiàn)卵巢靜止的奶牛血漿肌酸濃度降低和酪氨酸濃度升高的新變化。多種代謝異常提示奶牛卵巢靜止與泌乳早期能量負(fù)平衡有關(guān),并有伴發(fā)酮病和脂肪肝病的風(fēng)險(xiǎn);另外,卵巢靜止奶牛血漿肌酸、膽堿濃度降低和酪氨酸濃度升高的變化,為今后更為深入研究奶牛卵巢靜止發(fā)生機(jī)制提供了新的方向。
[Abstract]:For the study of ovarian quiescence and estrus in dairy cow dairy cows of different metabolites change, selection of postpartum 60~90d age, parity, body condition similar to healthy multiparous cows as experimental animal. According to the findings, oestrus rectal examination and ultrasound examination and hormone test results, select 10 cows for estrus group (A), 10 cows with ovarian quiescence group (B), then the two groups of cows plasma samples were unbiased 1 H NMR detection, the comparison of plasma generation metabolomics map and by multivariate statistical analysis, screening and identification of ovarian static cow plasma metabolite difference, and bioinformatics analysis. The results showed that: compared with normal estrous cows, ovarian static cow plasma metabolites of the 12 abnormalities, including increased levels of acetic acid, citric acid and tyrosine, low density lipoprotein, and decreased levels of very low density lipoprotein, lipid, alanine, pyruvic acid, creatine, choline, phosphocholine and glycerophosphocholine etc.; these metabolites and glucose metabolism, amino acid metabolism, lipid metabolism and choline metabolism is closely related to the abnormal regulation; through participation in the biosynthesis pathway, interferes with the normal follicular growth in dairy cows. And a new change in the decrease of plasma creatine concentration and the increase of tyrosine concentration in the plasma of the cows with the stillness of the ovaries was also found. A variety of metabolic abnormal ovarian quiescence in dairy cows in early lactation and negative energy balance, and is associated with ketosis and fatty liver disease risk; in addition, reduced ovarian static cow plasma creatine, choline concentration change and tyrosine concentration increased, for the future more in-depth study of the mechanism of ovary still provides a new direction.
