miR-20調(diào)控人乳牙牙髓干細(xì)胞向神經(jīng)元樣細(xì)胞的定向分化

背景:研究發(fā)現(xiàn)miR-20在促進(jìn)骨髓間充質(zhì)干細(xì)胞以及炎癥牙周膜干細(xì)胞的成骨分化中具有重要意義,但其在牙髓干細(xì)胞向神經(jīng)元細(xì)胞分化中的調(diào)節(jié)功能尚未完全闡明。目的:探討miR-20調(diào)控骨形態(tài)發(fā)生蛋白信號通路誘導(dǎo)人乳牙牙髓干細(xì)胞向神經(jīng)元樣細(xì)胞定向分化的機(jī)制。方法:從人脫落乳牙中分離純化乳牙牙髓干細(xì)胞并進(jìn)行神經(jīng)誘導(dǎo)分化,觀察誘導(dǎo)分化期間細(xì)胞形態(tài)變化,檢測Nestin、NSE、miR-20、BMP2的表達(dá)。將乳牙牙髓干細(xì)胞分為如下5組:陰性對照組(轉(zhuǎn)染陰性對照序列)、miR-20 mimic組(轉(zhuǎn)染mi R-20模擬物48 h)、mi R-20 inhibitor組(轉(zhuǎn)染miR-20抑制物48 h)、通路抑制劑組(骨形態(tài)發(fā)生蛋白通路抑制劑處理細(xì)胞10 d)、聯(lián)合組(轉(zhuǎn)染mi R-20模擬物并且在轉(zhuǎn)染后24 h在培養(yǎng)基中添加通路抑制劑處理細(xì)胞10 d)。各組乳牙牙髓干細(xì)胞在神經(jīng)誘導(dǎo)分化第12天,采用Real-time PCR和Western blot檢測BMP2、BMPR2、Nestin和NSE的m RNA和蛋白水平,免疫熒光染色觀察Ⅲβ-tubulin的表達(dá),高爾基染色檢測神經(jīng)元細(xì)胞樹突棘數(shù)目。結(jié)...
