骨形成蛋白7對大鼠不同部位脂肪源干細(xì)胞分化成棕色樣脂肪細(xì)胞的影響

目的探討骨形成蛋白7對大鼠不同部位脂肪組織脂肪源間充質(zhì)干細(xì)胞(AD-MSCs)誘導(dǎo)分化成棕色樣脂肪細(xì)胞的影響。方法分離大鼠背部肩胛間區(qū)棕色脂肪組織(i BAT)、雙側(cè)下肢腹股溝皮下白色脂肪組織(s WAT)和附睪白色脂肪組織(e WAT),分別提取脂肪源間充質(zhì)干細(xì)胞培養(yǎng),用骨形成蛋白7誘導(dǎo)干細(xì)胞分化。細(xì)胞免疫熒光染色、油紅O染色鑒定棕色樣脂肪細(xì)胞,PCR法檢測棕色脂肪相關(guān)基因表達(dá)。結(jié)果 i BAT和s WAT組誘導(dǎo)細(xì)胞中出現(xiàn)成簇聚集的空泡狀細(xì)胞,空泡狀細(xì)胞油紅O染色為紅色,解偶聯(lián)蛋白1免疫熒光染色陽性。e WAT組僅有散在的空泡狀細(xì)胞,解偶聯(lián)蛋白1免疫熒光染色陰性。反轉(zhuǎn)錄PCR顯示i BAT和s WAT組細(xì)胞高表達(dá)解偶聯(lián)蛋白1基因,未檢測到e WAT組細(xì)胞表達(dá)解偶聯(lián)蛋白1基因。結(jié)論大鼠體內(nèi)不同部位脂肪組織源干細(xì)胞的基因表達(dá)及分化潛能不同。
