人臍帶間充質(zhì)干細(xì)胞分離鑒定及向角質(zhì)形成細(xì)胞的誘導(dǎo)分化

該研究從人臍帶華通膠中獲得了人臍帶間充質(zhì)干細(xì)胞(human umbilical cord mesenchymal stem cells, hUCMSCs),通過基因鑒定和分化能力檢測(cè)其純度,建立hUCMSCs系。檢測(cè)結(jié)果表明,獲得的hUCMSCs表達(dá)轉(zhuǎn)錄因子SOX2、間充質(zhì)干細(xì)胞表面標(biāo)志物基因CD29、CD90和CD105,不表達(dá)造血系細(xì)胞表面標(biāo)志物基因CD34、CD45及人白細(xì)胞抗原基因HLA-DR。hUCMSCs可以向神經(jīng)細(xì)胞和成骨細(xì)胞分化,證明其多向分化潛能。P3代hUCMSCs向角質(zhì)細(xì)胞分化后,免疫熒光染色鑒定結(jié)果為陽性,實(shí)時(shí)熒光定量PCR檢測(cè)表達(dá)角蛋白CK19基因,隨著誘導(dǎo)分化時(shí)間的增加而升高。該實(shí)驗(yàn)建立了hUCMSCs系和角質(zhì)形成細(xì)胞的誘導(dǎo)分化體系,為后續(xù)研究奠定基礎(chǔ)。
