外周血有核細(xì)胞中tPA mRNA的表達(dá)與血栓性疾病的相關(guān)性研究
[Abstract]:Objective: thrombosis plays an important role in the occurrence of many cardiovascular events and is very harmful to human health. The timely and accurate diagnosis and thrombolysis effect of thrombus diseases play a key role in the prognosis of thrombus diseases. Tissue type PA (t-PA) plays an important role in human fibrinolytic system, and its abnormal activity is the most risk factor for prethrombus state in cardiovascular diseases. In this study, RT-PCR assay was used to detect the expression of t-PA mRNA in peripheral blood nucleated cells in thrombus group and prethrombus state group, which was significantly different from that in normal subjects. Methods: from October 2004 to December 2004, 40 patients with cerebral infarction were collected from Department of Neurology and Cardiovascular Medicine of Yantai Yuhuangding Hospital. At the same time, 20 patients with hypertension and 20 patients with coronary heart disease were taken. 40 cases of normal control were examined for health. The expression of t-PAmRNA in peripheral blood nucleated cells was detected by reverse transcription-PCR (RT-PCR), and the activity of t-PA in plasma was measured by chromogenic substrate method. Results: the expression of t-PA mRNA in peripheral blood nucleated cells in thrombus group and prethrombus state group was significantly higher than that in normal control group (P 0.01). The activity of t-PA in plasma of the two groups was also significantly decreased. Compared with the normal control group, there was significant difference (P 0.01). The expression of t-PA mRNA in peripheral blood nucleated cells is closely related to prethrombus state. The experimental results were misrepresented by mean 鹵standard, and statistically analyzed by Sigmastat 2.0 statistical software. The statistical methods used were t test and regression analysis. P05 and P01 showed that the difference was statistically significant and statistically significant. Conclusion: the expression of t-PA mRNA in peripheral blood nucleated cells is closely related to thrombosis and prethrombus state, and may be used as an early auxiliary monitoring index of prethrombus state.
