【摘要】：目的:血栓形成在許多心血管事件的發(fā)生中起著重要的作用,對(duì)人體健康危害極大。血栓性疾病的及時(shí)、準(zhǔn)確診斷及溶栓治療效果,對(duì)疾病的預(yù)后起著關(guān)鍵作用。組織型纖溶酶原激活物(t-PA)在人體纖溶系統(tǒng)中發(fā)揮著重要作用,其活性異常是心血管疾病血栓前狀態(tài)的最危險(xiǎn)因素。本實(shí)驗(yàn)采用RT-PCR法檢測(cè)血栓形成組及血栓前狀態(tài)組外周血有核細(xì)胞中t-PA mRNA的表達(dá)與正常人有無(wú)明顯差異。 方法:收取了煙臺(tái)毓璜頂醫(yī)院神經(jīng)內(nèi)科及心血管內(nèi)科自2004年10月至2004年12月收治的腦梗塞患者40例,采集外周血樣本。同期取高血壓患者20例,冠心病患者20例。正常對(duì)照40例,為健康查體者。分別用逆轉(zhuǎn)錄-PCR(RT-PCR)法檢測(cè)其外周血有核細(xì)胞中t-PAmRNA的表達(dá),發(fā)色底物法測(cè)定血漿t-PA的活性。 結(jié)果:血栓形成組及血栓前狀態(tài)組外周血有核細(xì)胞中t-PA mRNA的表達(dá)均明顯增強(qiáng),同正常照組比較有顯著性差異(P0.01),兩組人群血漿中t-PA的活性也都明顯降低,同正常對(duì)照組比較有顯著性差異(P0.01)。外周血有核細(xì)胞中t-PA mRNA的表達(dá)與血栓前狀態(tài)密切相關(guān)。實(shí)驗(yàn)結(jié)果采用均數(shù)±標(biāo)準(zhǔn)誤表示,用Sigmastat 2.0統(tǒng)計(jì)軟件進(jìn)行統(tǒng)計(jì)學(xué)分析,所用統(tǒng)計(jì)方法有t檢驗(yàn)、回歸分析。P0.05,P0.01分別表示差別具有統(tǒng)計(jì)學(xué)意義和顯著統(tǒng)計(jì)學(xué)意義。 結(jié)論:外周血有核細(xì)胞中t-PA mRNA的表達(dá)與血栓形成及血栓前狀態(tài)密切相關(guān),有可能作為血栓前狀態(tài)的早期輔助監(jiān)測(cè)指標(biāo)。
[Abstract]:Objective: thrombosis plays an important role in the occurrence of many cardiovascular events and is very harmful to human health. The timely and accurate diagnosis and thrombolysis effect of thrombus diseases play a key role in the prognosis of thrombus diseases. Tissue type PA (t-PA) plays an important role in human fibrinolytic system, and its abnormal activity is the most risk factor for prethrombus state in cardiovascular diseases. In this study, RT-PCR assay was used to detect the expression of t-PA mRNA in peripheral blood nucleated cells in thrombus group and prethrombus state group, which was significantly different from that in normal subjects. Methods: from October 2004 to December 2004, 40 patients with cerebral infarction were collected from Department of Neurology and Cardiovascular Medicine of Yantai Yuhuangding Hospital. At the same time, 20 patients with hypertension and 20 patients with coronary heart disease were taken. 40 cases of normal control were examined for health. The expression of t-PAmRNA in peripheral blood nucleated cells was detected by reverse transcription-PCR (RT-PCR), and the activity of t-PA in plasma was measured by chromogenic substrate method. Results: the expression of t-PA mRNA in peripheral blood nucleated cells in thrombus group and prethrombus state group was significantly higher than that in normal control group (P 0.01). The activity of t-PA in plasma of the two groups was also significantly decreased. Compared with the normal control group, there was significant difference (P 0.01). The expression of t-PA mRNA in peripheral blood nucleated cells is closely related to prethrombus state. The experimental results were misrepresented by mean 鹵standard, and statistically analyzed by Sigmastat 2.0 statistical software. The statistical methods used were t test and regression analysis. P05 and P01 showed that the difference was statistically significant and statistically significant. Conclusion: the expression of t-PA mRNA in peripheral blood nucleated cells is closely related to thrombosis and prethrombus state, and may be used as an early auxiliary monitoring index of prethrombus state.
