【摘要】:5-羥色胺(5-HT)是神經(jīng)系統(tǒng)內(nèi)一種重要的神經(jīng)遞質(zhì),具有多種復(fù)雜的功能,如調(diào)節(jié)運(yùn)動(dòng)、傷害性感受、攝食行為、體溫以及焦慮和抑郁等。5-HT通過(guò)與分布在中樞、周?chē)窠?jīng)系統(tǒng)中不同的5-HT受體相結(jié)合而發(fā)揮作用。近年來(lái)分子生物學(xué)的迅猛發(fā)展使得多種5-HT受體相繼克隆成功。根據(jù)受體的氨基酸序列、藥理學(xué)特性及細(xì)胞內(nèi)信號(hào)轉(zhuǎn)導(dǎo)機(jī)制等,可將5-HT受體分為7種類(lèi)型,即5-HT_(1-7)受體,又可進(jìn)一步劃分為14種亞型。除5-HT_3受體亞型為配體門(mén)控離子通道受體外,其它5-HT受體均為具有七次跨膜結(jié)構(gòu)的G蛋白耦聯(lián)受體。在傷害性信息的傳遞和調(diào)控過(guò)程中,5-HT具有雙重作用。一般認(rèn)為,5-HT在中樞主要發(fā)揮鎮(zhèn)痛作用,而在外周可以發(fā)揮致痛的作用。由于缺乏針對(duì)每種5-HT受體的特異性抗體、激動(dòng)劑和拮抗劑,對(duì)脊髓(SC)背角和背根神經(jīng)節(jié)(DRG)內(nèi)的5-HT受體在傷害性信息傳遞和調(diào)控中的作用尚未完全闡明。 神經(jīng)損傷所致病理性痛是臨床上常見(jiàn)的病理性痛之一,它給患者帶來(lái)極大的痛苦。對(duì)于其發(fā)病機(jī)制的探討有助于解除患者病癥,改善其生活質(zhì)量。目前對(duì)于5-HT這種重要的神經(jīng)遞質(zhì)在其中的作用研究甚少,同時(shí)由于存在著多種5-HT受體亞型,故在神經(jīng)病理性痛中何種受體亞型參與了
