晚期糖基化終產(chǎn)物單克隆抗體的制備

【摘要】: 研究目的:為建立檢測(cè)血清中晚期糖基化終末產(chǎn)物(AGEs)濃度的微量測(cè)定技術(shù),為糖尿病慢性并發(fā)癥的診斷、治療及預(yù)后評(píng)估提供依據(jù),本研究試圖制備抗人AGEs單克隆抗體。 研究方法:D-葡萄糖和人血清白蛋白(HSA)溶解于磷酸鹽緩沖液(PBS)中,37℃恒溫孵育12周,同時(shí)以不含D-葡萄糖的HSA為對(duì)照。12周后透析凝膠過濾純化,熒光測(cè)定、蛋白定量及聚丙烯酰胺凝膠電泳(SDS-PAGE)電泳鑒定。取100μg純化后的第一峰AGE-HSA加等體積福氏佐劑免疫Babl/c小鼠,共3次,用聚乙二醇(PEG)行細(xì)胞融合,用雜交瘤細(xì)胞選擇性培養(yǎng)基次黃嘌呤、氨基喋呤、胸腺嘧啶核苷(HAT)及次黃嘌呤、胸腺嘧啶核苷(HT)選擇性培養(yǎng),間接酶聯(lián)免疫吸附實(shí)驗(yàn)(ELISA)篩選培養(yǎng)上清,包被的抗原為純化后的第一峰AGE-HSA,選擇對(duì)AGEs陽(yáng)性,對(duì)HSA陰性的多克隆孔進(jìn)行亞克隆,間接ELISA篩選亞克隆板孔的細(xì)胞培養(yǎng)上清。收集陽(yáng)性單克隆細(xì)胞株,注射Babl/c小鼠制備腹水,經(jīng)固相化甘露聚糖結(jié)合蛋白(MBP)柱純化腹水。行亞型鑒定,并通過間接ELISA、western blot對(duì)抗體行特異性鑒定,并進(jìn)行染色體鑒定。 結(jié)果:D-葡萄糖和HSA孵育12周后,反應(yīng)液呈棕色,SDS-PAGE示分子量大于HSA的葡萄糖-HSA偶聯(lián)產(chǎn)物,熒光測(cè)定表明產(chǎn)物系A(chǔ)GEs。經(jīng)Sephadex G-200純化后,得到高分子量和低分子量的兩個(gè)蛋白峰,分別稱為第一峰和第二峰。第一峰作為免疫原,免疫Babl/c小鼠后血清滴度大于1:106,細(xì)胞融合后篩選,獲得陽(yáng)性多克隆,經(jīng)亞克隆,得到5株陽(yáng)性單克隆細(xì)胞株。染色體鑒定示其染色體為106條。擴(kuò)增5株細(xì)胞株,注射小鼠腹腔產(chǎn)生腹水,亞型鑒定為IgM型,抗體特異性鑒定示對(duì)AGE-HSA有特異性。 結(jié)論:獲得抗人AGE-HSA單克隆抗體細(xì)胞株5株,抗體亞型為IgM型。
