近日節(jié)律系統(tǒng)對藥物依賴和學習記憶的影響

【摘要】:目的: 本文從藥物依賴和學習記憶的角度,研究近日節(jié)律系統(tǒng)對高級神經活動的影響和作用。包括兩部分:1、近日節(jié)律基因mPerl與嗎啡依賴的關系,探討近日節(jié)律基因mPerl在藥物依賴過程中的作用和機制;2、不同光暗循環(huán)與學習記憶的關系,研究不同光暗循環(huán)對小鼠學習記憶的影響和機制。 方法: 實驗一:近日節(jié)律基因mperl在小鼠嗎啡依賴中的作用 動物選用BALB/C小鼠。設計并合成針對mPerl的“10-23”DNA核酶。以無DNA核酶活性的寡核苷酸對照,對同時嗎啡皮下注射和DNA核酶腦室注射的小鼠以及先嗎啡皮下注射后DNA核酶腦室注射的小鼠進行研究。通過條件性位置偏愛實驗,檢測小鼠對嗎啡的藥物依賴;以Western blot法,檢測小鼠全腦mPERl的表達;同時,體外實驗驗證DNA核酶對mPerlmRNA的切割效率。 實驗二:不同光暗循環(huán)對小鼠學習記憶的影響 動物選用ICR小鼠。小鼠在控制環(huán)境條件下,人工提供光線,3組的光暗循環(huán)分別為LD5/5、LD12/12和LD22/22,飼養(yǎng)6周。檢測指標包括:小鼠的活動性、血漿皮質激素(放射免疫法)和學習記憶能力(Morris水迷宮)。 結果: 實驗一:DNA核酶不但在體外能夠切割mPerl mRNA,而且在體
[Abstract]:Objective: to study the effect and effect of circadian rhythm system on advanced nerve activity from the point of view of drug dependence and learning and memory. It includes two parts: 1. The relationship between circadian rhythm gene mPerl and morphine dependence, to explore the role and mechanism of circadian rhythm gene mPerl in the process of drug dependence; 2. The relationship between different light and dark cycles and learning and memory was studied, and the effect and mechanism of different light and dark cycles on learning and memory in mice were studied. Methods: experiment 1: the role of circadian rhythm gene mperl in morphine dependence in mice. BALB/C mice were selected. A "10 脳 23" DNA ribozyme for mPerl was designed and synthesized. Compared with oligodeoxynucleotides without DNA ribozyme activity, mice injected subcutaneously with morphine and intraventricular injection of DNA ribozyme and mice injected with morphine subcutaneous injection followed by intraventricular injection of DNA ribozyme were studied. The drug dependence of mice on morphine was detected by conditioned place preference test, the expression of mPERl in the whole brain of mice was detected by Western blot assay, and the cutting efficiency of DNA ribozyme on mPerlmRNA was verified in vitro. Experiment 2: effects of different light and dark cycles on learning and memory in mice ICR mice were selected. Under the control environment, the mice were artificially supplied with light, and the light and dark cycles of the three groups were fed with LD5/5,LD12/12 and LD22/22, for 6 weeks, respectively. The activity of mice, plasma corticosteroids (radioimmunity) and learning and memory ability (Morris water maze) were measured. Results: experiment 1: DNA ribozyme could not only cut mPerl mRNA, in vitro but also in vivo.
