【摘要】:全球HBV的感染者已超過(guò)3億,,人類迫切需要研究新型而有效治療乙肝的手段。乙型肝炎在臨床上的表現(xiàn)千變?nèi)f化,可導(dǎo)致患者機(jī)體嚴(yán)重?fù)p害,甚至危及生命,給臨床醫(yī)生的診斷和治療帶來(lái)極大的挑戰(zhàn)。為了準(zhǔn)確恰當(dāng)?shù)脑u(píng)估乙肝患者的病情,必須依賴相關(guān)的實(shí)驗(yàn)室檢測(cè)和輔助檢查,主要包括HBV血清標(biāo)志物檢測(cè)、肝臟酶類生化檢測(cè)、肝組織病理學(xué)等傳統(tǒng)實(shí)驗(yàn)室檢查方法以及B超、CT、MRI等彩像學(xué)檢查及胃鏡等器械檢查,隨著分子生物學(xué)和醫(yī)學(xué)免疫學(xué)的迅速發(fā)展以及對(duì)此疾病的更加深刻地認(rèn)識(shí),現(xiàn)在已存在建立更加敏感和早期的乙型病毒性肝炎的診斷和治療方法的可能。 乙型肝炎病毒多聚酶在整個(gè)復(fù)制過(guò)程中起著關(guān)鍵作用,包括正鏈DNA的修補(bǔ)、ε信號(hào)識(shí)別、蛋白引發(fā)、轉(zhuǎn)位、逆轉(zhuǎn)錄、DNA依賴的DNA聚合酶活性和RnaseH等活性。有學(xué)者推測(cè)急慢性乙肝感染中TP可能分別起不同作用。對(duì)TP功能分子機(jī)制的闡明,有可能提示決定HBV急性感染轉(zhuǎn)歸的關(guān)鍵因素。定性和定量地檢測(cè)TP在肝細(xì)胞中的表達(dá)以
[Abstract]:There are more than 300 million HBV infected people all over the world. It is urgent to study new and effective methods to treat hepatitis B. The clinical manifestation of hepatitis B is ever-changing, which can lead to serious damage to the patient body, even life-threatening, and bring great challenges to the diagnosis and treatment of clinicians. In order to accurately and properly evaluate the condition of hepatitis B patients, we must rely on the relevant laboratory tests and auxiliary tests, mainly including the detection of HBV serum markers, liver enzymes biochemical test, With the rapid development of molecular biology and medical immunology and the deeper understanding of the disease, traditional laboratory examination methods such as liver histopathology, B-ultrasound, CT,MRI and other imaging examinations, and gastroscopy and other instruments, have developed rapidly with the rapid development of molecular biology and medical immunology. It is now possible to establish more sensitive and early methods for diagnosis and treatment of hepatitis B. Hepatitis B virus polymerase plays a key role in the whole replication process, including positive chain DNA repair, 蔚 signal recognition, protein initiation, translocation, reverse transcription, DNA-dependent DNA polymerase activity and RnaseH activity. Some scholars speculate that TP may play a different role in acute and chronic hepatitis B infection. The elucidation of the functional molecular mechanism of TP may suggest the key factors that determine the outcome of acute HBV infection. The expression of TP in hepatocytes was detected qualitatively and quantitatively.
