可分泌高活性β-半乳糖苷酶的重組乳酸乳球菌株的構(gòu)建及其產(chǎn)酶特性的初步研究

【摘要】:目的 以高產(chǎn)β-半乳糖苷酶(β-galactosidase)的保加利亞乳桿菌(Lactobacillus bulgaricus)β-半乳糖苷酶基因(lacZ)為初始基因,遞歸PCR合成的乳酸乳球菌基因usp45為分泌信號(hào)肽基因,表達(dá)型質(zhì)粒pMG36e為載體,構(gòu)建重組質(zhì)粒pMG36e-lacZ和pMG36e-usp-lacZ。電轉(zhuǎn)化乳酸乳球菌MG1363,獲得能分泌高活性β-半乳糖苷酶的重組乳酸乳球菌,并初步研究其產(chǎn)酶特性。為進(jìn)一步構(gòu)建食品級(jí)重組乳酸桿菌,用于乳糖酶缺乏的治療和研究奠定基礎(chǔ)。 方法 本研究分5個(gè)部分: 1.β-半乳糖苷酶基因初始菌的鑒定及其產(chǎn)酶特性研究:對(duì)基因初
[Abstract]:Objective to use the 尾-galactosidase (尾-galactosidase)-producing (Lactobacillus bulgaricus) 尾-galactosidase gene (lacZ) as the initial gene of Lactobacillus bulgaricus. The gene usp45 of Lactococcus lactis synthesized by recursive PCR was used as the secretory signal peptide gene, and the expression plasmid pMG36e was used as the vector. The recombinant plasmids pMG36e-lacZ and pMG36e-usp-lacZ. were constructed. The recombinant lactococcus, which can secrete high activity 尾-galactosidase, was obtained by electrotransformation of Lactococcus lactis MG1363,. It lays a foundation for the further construction of food-grade recombinant lactobacillus for the treatment and research of lactase deficiency. Methods this study was divided into five parts: 1.
