P2X受體對豚鼠胃竇環(huán)行肌自發(fā)性收縮運動的影響

【摘要】:目的:探討P2X受體激動劑α,β-methylene ATP (α,β-MeATP)對豚鼠胃竇環(huán)行肌運動的影響及其離子通道機制.方法:采用EWG/B豚鼠,制備去黏膜胃竇環(huán)行肌條(10×1.5 mm),并將其固定于恒溫灌流槽內(nèi)(37℃),用碳酸氫鈉緩沖液連續(xù)灌流并通氧(950 mL/L O_2,50 mL/L CO_2).利用SMUP-E生物信號處理系統(tǒng)記錄胃竇平滑肌自發(fā)性收縮活動,Ⅱ型膠原酶消化法分離豚鼠胃竇環(huán)行肌單細胞,傳統(tǒng)全細胞膜片鉗技術記錄急性分離的胃竇平滑肌細胞膜電位和離子電流,包括鈣激活鉀電流、延遲整流型鉀電流及電壓依賴性鈣電流.結果:嘌呤能P2X受體激動劑α,β-MeATP明顯抑制豚鼠胃竇環(huán)行肌自發(fā)性收縮,并有明顯的量效倚賴關系:5,10,20,40,100μmol/Lα,β-MeATP作用下,環(huán)行肌收縮幅度分別由對照組的100%下降到90%±2%.81%±4%,68%±4%,59%±7%和29%±4%(P0.05).用神經(jīng)阻斷劑Tetrodotoxin (TTX)預處理后,α,β-MeATP對胃竇環(huán)行肌自發(fā)性收縮的抑制作用仍不受影響;在傳統(tǒng)全細胞膜片鉗電流鉗模式下,500μmol/Lα,β-MeATP不影響細胞膜電位,也對兩種外向鉀電流,即延遲整流型鉀電流和鈣激活鉀電流沒有影響;不同濃度的α,β-MeATP對電壓依賴性鈣電流沒有影響.結論:P2X受體激動劑抑制豚鼠胃竇環(huán)行肌自發(fā)性收縮,其作用機制不依賴內(nèi)在神經(jīng),也不依賴細胞膜離子通道以及膜電位改變.
