青春雙歧桿菌對(duì)Ⅰ型變態(tài)反應(yīng)的影響

【摘要】: 目的觀察青春雙歧桿菌對(duì)Ⅰ型變態(tài)反應(yīng)的抑制作用。 方法1、大鼠實(shí)驗(yàn):1)以卵白蛋白為變應(yīng)原致敏大鼠,制備主動(dòng)致敏的腹腔肥大細(xì)胞。2)選用體外抗卵白蛋白主動(dòng)致敏的大鼠腹腔肥大細(xì)胞為細(xì)胞模型,分為實(shí)驗(yàn)組和模型組。分別進(jìn)行(1)甲苯胺藍(lán)染色法觀察青春雙歧桿菌對(duì)體外主動(dòng)致敏的肥大細(xì)胞脫顆粒的影響。(2)熒光分光光度法測(cè)定青春雙歧桿菌對(duì)體外主動(dòng)致敏的大鼠腹腔肥大細(xì)胞釋放組胺的影響。(3)免疫組化法檢測(cè)實(shí)驗(yàn)組和模型組中主動(dòng)致敏的肥大細(xì)胞中5-HT的表達(dá)。2、小鼠實(shí)驗(yàn):以卵白蛋白為變應(yīng)原,建立Ⅰ型變態(tài)反應(yīng)的實(shí)驗(yàn)動(dòng)物模型,并將模型動(dòng)物分為實(shí)驗(yàn)組和模型組,分別灌胃青春雙歧桿菌活菌液和生理鹽水,13天后,取靜脈血,用流式細(xì)胞儀檢測(cè)CD23的表達(dá)情況。 結(jié)果實(shí)驗(yàn)組和模型組中,肥大細(xì)胞脫顆粒的百分率分別是42.52±2.94%和90.89±1.47%,5-HT的染色積分分別是285.7±12.85和197.6±11.37,肥大細(xì)胞釋放組胺的量分別是1.09±0.01ng/ml和1.30±0.06ng/ml,CD23的百分率分別是17.71±2.47%和27.44±2.49%,實(shí)驗(yàn)組和模型組相比有顯著性差異(P<0.05)。光鏡下觀察:實(shí)驗(yàn)組細(xì)胞形態(tài)規(guī)則、脫顆粒細(xì)胞數(shù)減少。 結(jié)論雙歧桿菌具有抑制肥大細(xì)胞脫顆粒的作用,抑制IgE的產(chǎn)生,從而減少Ⅰ型變態(tài)反應(yīng)的發(fā)生。
[Abstract]:Objective to observe the inhibitory effect of Bifidobacterium pubertifolia on type I allergy. Methods 1, rat experiment: 1) using ovalbumin as allergen to sensitize rat peritoneal mast cells, and 2) using rat peritoneal mast cells which were anti-ovalbumin sensitized in vitro as cell model. It was divided into experimental group and model group. (1) toluidine blue staining method was used to observe the effect of bifidobacterium pubertifolia on degranulation of mast cells sensitized in vitro. (2) fluorescence spectrophotometry was used to determine the effect of bifidobacterium pubertifolia on the active sensitization of rat abdomen in vitro. (3) Immunohistochemical method was used to detect the expression of 5-HT in activated mast cells in experimental group and model group. Mouse experiment: using ovalbumin as allergen, the experimental animal model of type I allergic reaction was established, and the model animals were divided into experimental group and model group. Bifidobacterium puberty bifidobacterium solution and normal saline were perfused by stomach respectively. After 13 days, venous blood was taken. The expression of CD23 was detected by flow cytometry. Results the percentage of mast cell degranulation in experimental group and model group was 42.52 鹵2.94% and 90.89 鹵1.47 respectively. The staining scores of 5-HT were 285.7 鹵12.85 and 197.6 鹵11.37, respectively. The percentage of histamine released by mast cells was 1.09 鹵0.01ng/ml and 1.30 鹵0.06 ng / ml CD23, respectively, which were 17.71 鹵2.47% and 27.44 鹵2.49%, respectively. There was a significant difference between the experimental group and the model group (P < 0. 05). Under the light microscope, the cells in the experimental group were regular in shape and the number of degranulated cells was decreased. Conclusion Bifidobacterium can inhibit the degranulation of mast cells and the production of IgE, thus reducing the occurrence of type I allergic reaction.
