【摘要】： 目的:探討一氧化氮合酶和雌激素受體在下丘腦諸核團的分布及共存,為揭示雌激素與一氧化氮之間內(nèi)在聯(lián)系提供形態(tài)學證據(jù)。方法:對30只SD雌性大鼠下丘腦切片,采用還原型尼克酰胺腺嘌呤二核苷酸脫氫酶(NADPH-d)組織化學方法并結(jié)合免疫組織化學技術(shù),觀察大鼠下丘腦內(nèi)NOS陽性神經(jīng)元、ER陽性神經(jīng)元以及NOS/ER雙染神經(jīng)元的形態(tài)及分布。同時結(jié)合圖像分析技術(shù),計算下丘腦各核團內(nèi)NOS陽性神經(jīng)元、ER陽性神經(jīng)元以及NOS/ER雙染神經(jīng)元的數(shù)目和雙染神經(jīng)元所占的比例。結(jié)果:①NOS陽性神經(jīng)元主要分布在下丘腦室旁核、視上核、下丘腦外側(cè)區(qū)和室周核。②ER陽性神經(jīng)元在下丘腦諸核團的表達不及NOS陽性神經(jīng)元廣泛,主要分布在下丘腦視上核、室旁核和下丘腦外側(cè)區(qū),此外在室周核和大腦皮層也見到少量散在分布的ER陽性神經(jīng)元。③NOS與ER雙染神經(jīng)元主要分布在下丘腦的室旁核、視上核、下丘腦外側(cè)區(qū)及室周核。其它區(qū)域可見到散在分布的雙染神經(jīng)元。視上核雙染神經(jīng)元主要位于視上核背內(nèi)側(cè)和背外側(cè)部。室旁核的雙染神經(jīng)元主要分布在小細胞部的腹內(nèi)側(cè)區(qū)。在下丘腦外側(cè)區(qū),雙染神經(jīng)元分布較廣。室周核可見零星分布的雙染神經(jīng)元。結(jié)論:1在大鼠下丘腦視上核、室旁核、下丘腦外側(cè)區(qū)及室周核均有NOS與ER雙染神經(jīng)元分布。2 NOS與ER雙染神經(jīng)元主要集中分布在視上核的背內(nèi)側(cè)和背外側(cè)部及室旁核小細胞部腹內(nèi)側(cè)區(qū),在下丘腦外側(cè)區(qū)分布較廣但比較分散,室周核呈散在分布。
[Abstract]:Aim: to investigate the distribution and coexistence of nitric oxide synthase (NOS) and estrogen receptor (ER) in hypothalamic nuclei, and to provide morphological evidence for revealing the relationship between estrogen and nitric oxide (no). Methods: the NOS positive neurons in the hypothalamus of 30 SD female rats were observed by using reduced nicotinamide adenine dinucleotide dehydrogenase (NADPH-d) histochemistry and immunohistochemical technique. Morphology and distribution of ER positive neurons and NOS/ER double staining neurons. At the same time, the number and proportion of NOS positive neurons, ER positive neurons and NOS/ER double staining neurons in each nucleus of hypothalamus were calculated with image analysis technique. Results: 1NOS positive neurons were mainly distributed in the paraventricular nucleus of hypothalamus, supraoptic nucleus, lateral hypothalamic area and periventricular nucleus. The expression of 2ER positive neurons in the nuclei of hypothalamus was not as wide as that of NOS positive neurons, but mainly in the supraoptic nucleus of hypothalamus. The paraventricular nucleus and the lateral hypothalamus were also found in the paraventricular nucleus and lateral hypothalamus, and a few scattered ER positive neurons were found in the periventricular nucleus and cerebral cortex. The double staining neurons of 3NOS and ER were mainly distributed in the paraventricular nucleus, supraoptic nucleus, lateral hypothalamic area and periventricular nucleus of the hypothalamus. Scattered double-stained neurons can be seen in other regions. The double staining neurons of supraoptic nucleus were mainly located in the dorsomedial and dorsolateral part of supraoptic nucleus. The double-staining neurons in the paraventricular nucleus were mainly located in the ventromedial region of the small cells. In the lateral hypothalamus, double staining neurons are widely distributed. Scattered double-stained neurons were found in the periventricular nucleus. Conclusion: 1 in the supraoptic nucleus and paraventricular nucleus of the hypothalamus of rats, The distribution of NOS and ER double staining neurons in lateral hypothalamic area and periventricular nucleus. 2 NOS and ER double staining neurons were mainly distributed in dorsomedial and dorsolateral part of supraoptic nucleus and ventromedial area of small cell part of paraventricular nucleus. The lateral hypothalamus was widely distributed, and the periventricular nucleus was scattered.
