聯(lián)合轉(zhuǎn)染P~(21)基因和反義c-fos核酸對自體移植靜脈中膜平滑肌細胞增生的影響

【摘要】:目的:本工作通過觀察聯(lián)合轉(zhuǎn)染P~(21)基因和反義c-fos核酸對自體移植靜脈的中膜平滑肌細胞(VSMC)增生的影響,探索一種理想的抑制自體移植血管再狹窄的基因療法。 方法:選擇健康雄性新西蘭家兔20只,隨機分成實驗和對照組兩組,每組各10只,所有動物均行自體右頸靜脈和右頸總動脈移植手術(shù)。實驗組移植靜脈以腺病毒介導的P~(21)cDNA溶液浸泡和反義c-fos寡聚核苷酸凝膠涂布;對照組僅行空載腺病毒溶液浸泡和凝涂布。術(shù)后2周取出移植血管,分別對移植血管中膜平滑肌細胞進行計數(shù)和對移植血管中膜平滑肌細胞增殖細胞核抗原(PCNA)進行測定,測定PCNA陽性表達和VSMC的DNA含量及管腔狹窄度。 結(jié)果:實驗組動物的VSMC增殖指數(shù)為12.74±1.78,對照組為26.25±5.75兩組比較有極顯著差異(P0.01);實驗組PCNA陽性表達率為32.90±3.64,對照組為55.83±3.92兩組比較有極顯著差異(P0.01);實驗組VSMC的DNA含量為5.79±1.59,對照組為11.62±2.19兩組比較有極顯著差異(P0.01);實驗組管腔狹窄度為11.50±1.85,對照組為23.34±5.79兩組比較有極顯著差異(P0.01)。 結(jié)論:①聯(lián)合轉(zhuǎn)染P~(21)和反義c-fos可有效抑制自體移植靜脈中膜平滑肌細胞的增生。②聯(lián)合轉(zhuǎn)染P~(21)和反義c-fos是一種有效的防治移植靜脈發(fā)生再狹窄的基因療法。
[Abstract]:Aim: to investigate the effect of combined transfection of P21 gene and antisense c-fos nucleic acid on the proliferation of (VSMC) in medial smooth muscle cells (MSMCs) after autologous vein transplantation, and to explore an ideal gene therapy for inhibiting restenosis of autologous grafts. Methods: twenty healthy male New Zealand rabbits were randomly divided into experimental group and control group with 10 rabbits in each group. Autogenous right jugular vein and right common carotid artery transplantation were performed in all the rabbits. The vein grafts in the experimental group were immersed in adenovirus-mediated P21 cDNA solution and coated with antisense c-fos oligonucleotide gel while those in the control group were only soaked in empty adenovirus solution and coated with coagulant. The vascular grafts were removed 2 weeks after operation. The number of MSMCs and the proliferating cell nuclear antigen (PCNA) were measured respectively. The positive expression of PCNA, the DNA content of VSMC and the degree of lumen stenosis were measured. Results: the proliferative index of VSMC was 12.74 鹵1.78 in the experimental group and 26.25 鹵5.75 in the control group (P0.01). The positive expression rate of PCNA was 32.90 鹵3.64 in the experimental group and 55.83 鹵3.92 in the control group (P0.01). The DNA content of VSMC was 5.79 鹵1.59 in the experimental group and 11.62 鹵2.19 in the control group (P0.01). The degree of lumen stenosis was 11.50 鹵1.85 in the experimental group and 23.34 鹵5.79 in the control group (P0.01). Conclusion: 1 Co-transfection of P21 and antisense c-fos can effectively inhibit proliferation of MSMCs in autogenous vein grafts. 2 Co-transfection of P21 and antisense c-fos is an effective method to prevent and treat vein graft reoccurrence.
