可視法腦片膜片鉗實(shí)驗(yàn)技術(shù)的建立及大鼠前庭內(nèi)側(cè)核神經(jīng)元毒蕈堿樣膽堿能受體電生理特性的研究

【摘要】: 【目的】建立大鼠前庭內(nèi)側(cè)核腦片可視法膜片鉗實(shí)驗(yàn)技術(shù),探討在前庭內(nèi)側(cè)核神經(jīng)元(medial vestibular nucleus neurons,MVNn)中毒蕈堿樣膽堿能受體(M受體)介導(dǎo)電流的特性。 【方法】應(yīng)用紅外微分干涉相差(infrared differential interference contrast,IR-DIC)顯微鏡結(jié)合電荷耦合式攝像機(jī)(charge coupled device-camera,CCD-camera)系統(tǒng),可視法膜片鉗全細(xì)胞記錄模式下檢測前庭內(nèi)側(cè)核神經(jīng)元毒蕈堿樣膽堿能受體介導(dǎo)電流的特性。 【結(jié)果】可視法腦片膜片鉗技術(shù)可對神經(jīng)元直接進(jìn)行準(zhǔn)確定位以及健康細(xì)胞的篩選。在MVNn中,毒蕈堿引發(fā)的M受體去極化效應(yīng)是由鉀電導(dǎo)的減少所介導(dǎo),并證明所抑制的離子電流是漏鉀電流。 【結(jié)論】可視法腦片膜片鉗實(shí)驗(yàn)技術(shù)克服了盲法腦片膜片鉗技術(shù)的缺陷,大大提高了對神經(jīng)元封接的成功率。通過對MNVn中M受體通道電流性質(zhì)的分析,從而進(jìn)一步揭示毒蕈堿樣膽堿能機(jī)制在MVNn的興奮性調(diào)節(jié)作用,并為臨床抗膽堿藥物的應(yīng)用提供新的思路。
[Abstract]:[objective] to establish a visual patch clamp technique in the medial vestibular nucleus of rats to investigate the characteristics of muscarinic cholinergic receptor (M receptor) mediated currents in the neurons of the medial vestibular nucleus (medial vestibular nucleus neurons,MVNn). [methods] an infrared differential interference phase difference (infrared differential interference contrast,IR-DIC) microscope combined with a charge-coupled camera (charge coupled device-camera,CCD-camera) system was used. The characteristics of muscarinic cholinergic receptor-mediated currents in medial vestibular nucleus neurons were detected by visual patch clamp whole-cell recording mode. [results] the visual patch clamp technique can directly locate neurons and screen healthy cells. In MVNn, the depolarization of M receptor induced by muscarinic acid is mediated by the reduction of potassium conductance, and it is proved that the inhibited ion current is potassium leakage current. [conclusion] the visual patch-clamp technique overcomes the defect of blind patch-clamp technique and improves the success rate of neuron sealing. By analyzing the current properties of M-receptor channel in MNVn, the excitatory regulation of muscarinic cholinergic mechanism in MVNn was further revealed, and a new idea for clinical application of anticholine drugs was provided.
