結(jié)核分枝桿菌Rpf樣蛋白生物學(xué)及免疫學(xué)活性的研究

【摘要】:結(jié)核病(Tuberculosis,TB)是由結(jié)核分枝桿菌(Mycobacterium tuberculosis,MTB)引起的以呼吸道傳播為主的慢性傳染性疾病。近年來(lái),由于卡介苗(BCG)保護(hù)性的不完善、MTB耐藥性的產(chǎn)生、艾滋病的流行、世界人口的劇增、全球性旅游業(yè)的發(fā)展以及吸毒、酗酒、貧窮等原因,TB的發(fā)病率呈日益嚴(yán)重的回升趨勢(shì),再次成為危害人類(lèi)健康的世界性疾病。WHO指出,全世界現(xiàn)有TB患者2000萬(wàn),每年新發(fā)生約900萬(wàn)例,每年死亡約300萬(wàn)例。WHO預(yù)測(cè),在未來(lái)的10年中,全球?qū)⒂谐^(guò)3億人感染MTB,9000萬(wàn)人發(fā)病,3000萬(wàn)人死于TB。因此,積極研究TB的致病及免疫機(jī)制,研制更為有效的診斷、預(yù)防和治療MTB感染的新技術(shù)、新方法、新疫苗等具有重要意義。 藤黃微球菌(Micrococcus luteus)中的促進(jìn)復(fù)活因子(resuscitation-promoting factor,Rpf)是被發(fā)現(xiàn)的第一種細(xì)菌的細(xì)胞因子,它具有促進(jìn)休眠菌的復(fù)蘇以及促進(jìn)細(xì)菌快速生長(zhǎng)的作用。序列對(duì)比分析發(fā)現(xiàn),在其他的一些革蘭陽(yáng)性細(xì)菌如MTB、麻風(fēng)分枝桿菌、藍(lán)色鏈霉菌、谷氨酸棒桿菌等中也存在著30多個(gè)與Rpf同源的蛋白,其中在MTB中就有5個(gè)Rpf樣蛋白,分別為Rv1884c、Rv2389c、Rv2450、Rv1009和Rv0867,推測(cè)這些蛋白也可能具有促進(jìn)MTB快速生長(zhǎng)的作用。此外,
