STAg聯(lián)合IFN-γ和蜂膠鼻內(nèi)免疫小鼠誘導(dǎo)的抗弓形蟲感染保護(hù)性免疫應(yīng)答

【摘要】:目的本研究首先探討STAg 分別聯(lián)合不同劑量IFN-γ、蜂膠鼻內(nèi)免疫對(duì)小鼠的保護(hù)作用及IFN-γ、蜂膠作為佐劑鼻內(nèi)免疫抗弓形蟲感染的最佳劑量。再比較IFN-γ、蜂膠兩種佐劑最佳劑量以及兩種佐劑混合鼻內(nèi)免疫輔助STAg增強(qiáng)機(jī)體抗弓形蟲攻擊的能力,并探索兩種佐劑聯(lián)合應(yīng)用的免疫效果。 方法確定IFN-γ、蜂膠最佳劑量的實(shí)驗(yàn)方法:分別將70 只、60 只5~6 周齡BALB/c小鼠隨機(jī)分為5個(gè)組,用20μg STAg分別聯(lián)合不同劑量IFN-(γ250U,500U,1000U,2000U)、蜂膠(5μg,10μg,20μg, 40μg)鼻內(nèi)免疫小鼠2 次,間隔14 天,末次免疫后第10 天,用RH 株弓形蟲速殖子4×104 個(gè)/只灌胃攻擊,逐日觀察小鼠健康存活情況。攻擊后第29 天處死全部小鼠,分離計(jì)數(shù)腦、肝組織速殖子;計(jì)數(shù)腸系膜淋巴結(jié)(MLN)和脾T 淋巴細(xì)胞;ELISA 法檢測(cè)小鼠糞便、肺沖洗液弓形蟲特異性IgA 和血清IgG 抗體水平。 比較IFN-γ、蜂膠佐劑作用的實(shí)驗(yàn)方法:將5~6 周齡雌性BALB/c 小鼠60 只隨機(jī)分為4 個(gè)組,每組15 只,分別用20μg STAg、20μg STAg+40μg蜂膠、20μg STAg+1000U IFN-γ以及20μg STAg+40μg 蜂膠+1000U IFN-γ鼻內(nèi)免疫小鼠2 次,間隔14 天,末次免疫后第10 天,用RH 株弓形蟲速殖子4×104 個(gè)/只灌胃攻擊,逐日記錄小鼠體重,觀察小鼠存活情況。攻擊后第43 天處死全部小鼠,分離計(jì)數(shù)腦、肝組織速殖子;計(jì)數(shù)IEL、PP 結(jié)和脾T 淋巴細(xì)胞,免疫細(xì)胞化學(xué)(ICC)法檢測(cè)其T 細(xì)胞亞群水平;ELISA 法檢測(cè)小鼠糞便、鼻咽沖洗液、肺沖洗液、陰道沖洗液和血清中弓形蟲特異性IgA 和血清IgG 抗體水平。 結(jié)果隨佐劑IFN-γ劑量的增加,小鼠存活率有升高趨勢(shì),1000U IFN-γ劑量組小鼠存活率最高達(dá)93%;腦、肝速殖子數(shù)呈下降的趨勢(shì), 其中STAg
[Abstract]:Objective to investigate the protective effect of STAg combined with different doses of IFN- 緯, propolis nasal immunization on mice and the optimal dose of IFN- 緯 and propolis as adjuvant in nasal immunity against Toxoplasma gondii infection. The best dosage of IFN- 緯, two adjuvants of propolis and two adjuvants combined with STAg were compared to enhance the ability of body to resist Toxoplasma gondii attack, and to explore the immune effect of the two adjuvants combined with the two adjuvants.
