口蹄疫重組蛋白疫苗的計算機(jī)輔助設(shè)計及功能研究

【摘要】: 口蹄疫( Foot and mouth disease , FMD )是由口蹄疫病毒(Foot-and-mouth disease virus,FMDV)引起的一種急性高度傳染性的疾病。由于其具有高度傳染性和對農(nóng)業(yè)經(jīng)濟(jì)的重大影響,國際獸醫(yī)局(OIE)將口蹄疫列為A類傳染病。 雖然接種口蹄疫滅活疫苗是預(yù)防和控制FMD爆發(fā)流行的有效手段,但因其在生產(chǎn)時易造成病毒泄漏,病毒滅活不全,有病毒擴(kuò)散的隱患,促使我們尋求一種安全有效的疫苗來替代它,重組蛋白疫苗無疑是很好選擇。 隨著計算機(jī)技術(shù)的發(fā)展,表位預(yù)測、蛋白質(zhì)結(jié)構(gòu)預(yù)測使疫苗的設(shè)計有據(jù)可循。我們運用計算機(jī)技術(shù)設(shè)計了AsiaⅠ型口蹄疫疫苗RA2,并用基因工程的手段構(gòu)建了重組牛AsiaⅠ型口蹄疫疫苗RA2的編碼基因,在大腸桿菌中表達(dá),并檢驗了該重組疫苗的免疫效果。 免疫豚鼠的實驗表明,RA2疫苗在豚鼠體內(nèi)可誘生高水平的抗AsiaⅠFMDV的中和性抗體。攻毒實驗表明,RA2疫苗免疫的牛有3/5得到了保護(hù),提示RA2可能成為抗牛AsiaⅠ型口蹄疫病毒的有效疫苗。 綜上所述,采用計算機(jī)輔助設(shè)計,可以以較少的工作量構(gòu)建出免疫效果較好的疫苗。

Although vaccination with inactivated vaccine against foot-and-mouth disease is an effective means to prevent and control the outbreak of FMD, it is easy to cause virus leakage and inactivation of the virus during production. In order to find a safe and effective vaccine to replace it, recombinant protein vaccine is undoubtedly a good choice.
