大鼠前列腺傷害性刺激時脊髓、孤束核P2X3受體、Fos的表達(dá)

【摘要】:目的:研究大鼠前列腺傷害性刺激與P2X3受體、Fos在脊髓內(nèi)、孤束核的表達(dá)及其意義。方法:建立大鼠前列腺傷害性刺激動物模型,按完全隨機(jī)化設(shè)計,三因素三水平析因試驗,因素水平共有27種組合,健康雄性Wistar大鼠54只隨機(jī)分為27組,每組2只,分別給予上述27種處理。如27號鼠接受三種因素的第三水平處理,即:腹正中切口,剪斷腹下叢;腹正中切口,直視下向前列腺兩葉各注射大腸桿菌(0∶1H∶2Z17)懸液0.1ml(濃度1×10~8個/ml);干預(yù)后存活72小時。余者類推。用抗Fos蛋白和抗P2X3受體的免疫組織化學(xué)方法,對大鼠前列腺傷害性刺激后脊髓內(nèi)、孤束核內(nèi)Fos、P2X3受體免疫陽性物表達(dá)部位進(jìn)行觀察。結(jié)果:在脊髓L_1水平切面上,可見Fos免疫陽性細(xì)胞主要分布在脊髓背角Ⅰ、Ⅱ板層,后聯(lián)合、中央管周圍和前角也有分布。P2X3受體陽性細(xì)胞主要見于脊髓背角Ⅱ板層。孤束核有不同程度的Fos、P2X3受體免疫陽性細(xì)胞表達(dá)(P0.05)。前列腺的交感通路切斷后,L_1水平的Fos、P2X3受體免疫陽性細(xì)胞數(shù)明顯下降(P0.05)。2%甲醛液引起的P2X3受體免疫陽性細(xì)胞數(shù)比大腸桿菌引起的多(P0.05)。結(jié)論:交感神經(jīng)通路在大鼠前列腺傷害性信息傳遞過程中具有重要作用;P2X3受體可能參與了前列腺傷害性刺激的傳導(dǎo)調(diào)制;孤束核可能參與了前列腺傷害性刺激信息的調(diào)制。
[Abstract]:Aim: to study the expression and significance of nociceptive stimulation and P2X3 receptor Fos in spinal cord of rats. Methods: an animal model of noxious prostatic stimulation was established in rats. According to the totally randomized design, three factors and three levels of factorial test, there were 27 combinations at the factor level. 54 healthy male Wistar rats were randomly divided into 27 groups with 2 rats in each group. 27 treatments were given respectively. For example, 27 rats were treated at the third level of three kinds of factors, that is, median abdominal incision, cut off the lower abdominal plexus, median abdominal incision, injection of Escherichia coli (0:1H:2Z17) suspension 0.1ml (1 脳 108 / ml) into each of the two lobes of prostate directly, and survived 72 hours after intervention. The rest are analogies. The immunoreactive sites of Fos,P2X3 receptors in the spinal cord and nucleus tractus solitarius after noxious stimulation of rat prostate were observed by immunohistochemical method of anti Fos protein and anti P2X3 receptor.
