【摘要】：目的觀察復方鹿角霜對小鼠膠原誘導性關節(jié)炎(CIA)體內(nèi)相關炎癥細胞因子的表達及滑膜組織病理學的影響。方法建立小鼠CIA模型,實驗小鼠隨機分為5組:空白對照組,模型對照組,復方鹿角霜總膏低、高劑量組(C1,C2組),雷公藤多苷片組。小鼠第一次免疫后,觀察小鼠的一般情況,記錄小鼠前后足爪病變發(fā)生情況。小鼠第二次免疫后,復方鹿角霜C1、C2組分別灌胃給予復方鹿角霜總膏2.5,5 g·kg-1·d-1,雷公藤多苷片組灌胃給予雷公藤多苷片13.6 g·kg-1·d-1,連續(xù)4周。小鼠給藥結(jié)束后,取血分離血清,酶聯(lián)免疫吸附試驗(ELISA)檢測小鼠血清腫瘤壞死因子-α(TNF-α)和白細胞介素-4(IL-4)的表達水平。取小鼠足踝關節(jié),光鏡觀察關節(jié)滑膜組織病理學變化。結(jié)果與模型對照組比較,復方鹿角霜C1、C2組、雷公藤多苷片組血清TNF-α均明顯降低;而復方鹿角霜組血清IL-4水平升高。模型對照組關節(jié)病理可見滑膜組織增生,各給藥組關節(jié)病理形態(tài)有所改善。結(jié)論復方鹿角霜可減輕膠原誘發(fā)的小鼠關節(jié)炎紅腫癥狀,其作用機制可能與降低炎性因子TNF-α、調(diào)節(jié)抑炎因子IL-4作用有關。
[Abstract]:Objective to observe the effect of compound Lujiao cream on the expression of inflammatory cytokines and histopathology of synovium in (CIA) of mice with collagen-induced arthritis. Methods CIA model was established in mice. The experimental mice were randomly divided into 5 groups: blank control group, model control group, compound Lujiao cream low, high dose group (C1C 2 group), Tripterygium wilfordii polyglycoside group. After the first immunization, the general situation of mice was observed and the occurrence of paw lesions before and after immunization was recorded. After the second immunization, the compound antler cream C _ 1C _ 2 group was given the compound antler cream 2.5g kg-1 d ~ (-1) by gavage, and the tripterygium wilfordii polyglycoside tablet 13.6 g kg-1 d ~ (-1) by gavage for 4 weeks. Serum samples were collected from mice after administration, and the expression of tumor necrosis factor- 偽 (TNF- 偽) and interleukin-4 (IL-4) in serum of mice were detected by Elisa (ELISA). The serum levels of tumor necrosis factor 偽 (TNF- 偽) and interleukin-4 (IL-4) in mice were determined by enzyme-linked immunosorbent assay. The histopathological changes of synovial membrane were observed under light microscope. Results compared with the model control group, the serum TNF- 偽 of the C _ 1C _ 2 group and the Tripterygium wilfordii polyglycoside group were significantly decreased, while the serum IL-4 level of the compound antler cream group was higher than that of the control group. Synovial tissue proliferation was observed in the model control group, and the joint morphology was improved in each administration group. Conclusion compound Lujiao cream can alleviate the symptoms of arthritis redness and swelling induced by collagen in mice, and its mechanism may be related to the reduction of inflammatory factor TNF- 偽 and the regulation of anti-inflammatory factor IL-4.
