巴馬壯族長壽人群轉(zhuǎn)化生長因子-β1基因多態(tài)性的研究

【摘要】: 目的:探討轉(zhuǎn)化生長因子-β1(TGF-β1)基因的多態(tài)性與人類長壽的相關(guān)性。方法:采用聚合酶鏈式反應(yīng)-限制性片段長度多態(tài)性(PCR-RFLP)技術(shù),檢測132例廣西巴馬地區(qū)健康壯族長壽老人TGF-β1的基因多態(tài)性,包括TGF-β1基因啟動子區(qū)域G-800A、C-509T和第一外顯子T+869C、G+915C四個多態(tài)位點,并與118例巴馬地區(qū)健康壯族成年人和70例廣西賀州地區(qū)健康壯族人群作比較。結(jié)果:1.TGF-β1基因的G-800A、G+915C兩個多態(tài)位點在廣西巴馬地區(qū)及賀州地區(qū)未檢測出多態(tài)性;2.TGF-β1基因的C-509T、T+869C兩個多態(tài)位點的等位基因頻率和基因型頻率分布比較分析顯示,巴馬長壽組與對照組之間無顯著性差異(P>0.05),而巴馬長壽組與賀州壯族組(P_(-509)~(allele)=0.000,P_(-509)~(genotype)=0.000,P_(+869)~(allele)=0.000,P_(+869)~(genotype)=0.003)、巴馬對照組與賀州壯族組(P_(-509)~(allele)=0.034,P_(-509)~(genotype)=0.039,P_(+869)~(allele)=0.017,P_(+869)~(genotype)=0.026)之間均有顯著性差異;3.進一步的基因型組合分析顯示,TGF-β1基因G-800A、C-509T、T+869C、G+915C四個多態(tài)位點基因型組合GG-CT-TT-GG在巴馬長壽老人組中的分布明顯高于巴馬對照組(P=0.000);基因型組合GG-CT-TC-GG和GG-TT-CC-GG在巴馬長壽組和賀州壯族組(P_(GG-CT-TC-GG)=0.002,P_(GG-TT-CC-GG)=0.002)、巴馬對照組和賀州壯族組(P_(GG-CT-TC-GG)=0.002,P_(GG-TT-CC-GG)=0.031)之間均存在顯著差異。結(jié)論:巴馬地區(qū)壯族長壽人群TGF-β1基因某些多態(tài)位點的等位基因頻率、基因型頻率及基因型組合頻率有異于普通人群,與該地區(qū)的長壽呈一定的相關(guān)性,提示巴馬地區(qū)長壽人群存在獨特的抗炎性衰老機制,從而在一定程度上體現(xiàn)出生存優(yōu)勢。
[Abstract]:Objective: to investigate the association between the polymorphism of transforming growth factor- 尾 1 (TGF- 尾 1) gene and longevity. Methods: polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the gene polymorphism of TGF- 尾 1 in 132 healthy Zhuang people in Guangxi. Four polymorphic loci including G-800AnC-509T and G915C in the promoter region of TGF- 尾 1 gene were compared with 118 healthy adults in Bama area and 70 healthy Zhuang people in Hezhou area of Guangxi. Results 1. Two polymorphic loci of TGF- 尾 1 gene G-800Agng G915C were not detected in Bama and Hezhou regions of Guangxi. 2.
