【摘要】：目的:建立一種猴體神經(jīng)毒力實(shí)驗(yàn)的組織病理學(xué)評(píng)價(jià)方法,用于疫苗猴體神經(jīng)毒力非臨床安全性評(píng)價(jià)。方法:通常采用猴丘腦內(nèi)和脊髓腰椎段注射給予疫苗。給藥結(jié)束后,猴體經(jīng)解剖,取材,脫水,包埋,切片,HE染色后在光鏡下觀察神經(jīng)組織病理學(xué)改變。病變按規(guī)定的神經(jīng)毒性診斷標(biāo)準(zhǔn)打分后用Ridit方法進(jìn)行組間比較檢驗(yàn),最終對(duì)所觀察到的神經(jīng)組織病變的統(tǒng)計(jì)結(jié)果進(jìn)行分析和評(píng)價(jià)。結(jié)果:本方法中,腦組織取材包括左右兩側(cè)大腦在內(nèi)的14個(gè)切面以及小腦、腦橋1個(gè)切面和延髓2個(gè)切面;脊髓取材包括頸髓5個(gè)切面、胸髓3個(gè)切面和腰髓6個(gè)切面。以上所取切面涵蓋了神經(jīng)組織的基本結(jié)構(gòu)功能區(qū)域,操作簡(jiǎn)單且實(shí)用。組織病理學(xué)檢查將神經(jīng)組織病變分類為神經(jīng)元變性、壞死,嗜神經(jīng)現(xiàn)象,神經(jīng)膠質(zhì)細(xì)胞增多,小膠質(zhì)細(xì)胞結(jié)節(jié),圍管浸潤(rùn)和針跡反應(yīng)等。這些病變按病變數(shù)目和病灶面積進(jìn)行等級(jí)劃分,從"0"分到"4"分共分為5個(gè)等級(jí)。結(jié)論:神經(jīng)系統(tǒng)的形態(tài)學(xué)評(píng)價(jià)要求病理實(shí)驗(yàn)人員熟悉大腦的結(jié)構(gòu)和功能,掌握解剖及病理制片技能。因此,神經(jīng)組織病理制片的科學(xué)性和一致性以及操作人員的技術(shù)水平影響神經(jīng)毒性組織病理學(xué)評(píng)價(jià)的可靠性和準(zhǔn)確性。本研究建立的神經(jīng)毒力組織病理學(xué)評(píng)價(jià)方法除適用于疫苗猴體神經(jīng)毒力非臨床安全性評(píng)價(jià)實(shí)驗(yàn)外,還可應(yīng)用于一般神經(jīng)毒性實(shí)驗(yàn)、神經(jīng)組織退行性疾病動(dòng)物模型以及腦、脊髓損傷動(dòng)物模型的評(píng)價(jià)中,是一項(xiàng)系統(tǒng)、規(guī)范、實(shí)用的神經(jīng)毒性組織病理學(xué)評(píng)價(jià)方法。
[Abstract]:Objective: to establish a method for histopathological evaluation of somatic neurovirulence in monkeys. Methods: the vaccine was usually injected into the thalamus and spinal cord of monkeys. After administration, the monkey body was dissected, sampled, dehydrated, embedded, stained with HE and observed the pathological changes of nerve tissue under light microscope. The pathological changes were graded according to the prescribed criteria of neurotoxicity and compared with each other by Ridit method. Finally, the statistical results of the observed neuropathy were analyzed and evaluated. Results: in this method, 14 sections and cerebellum, 1 pons and 2 medulla oblongata were selected from brain tissue, 5 sections from cervical spinal cord, 3 from thoracic spinal cord and 6 from lumbar spinal cord. The above section covers the basic structure and function area of neural tissue and is simple and practical. Histopathological examination classified neuropathy as neuronal degeneration, necrosis, neurophilic phenomenon, glial cell proliferation, microglial nodule, perivascular infiltration and Needle reaction. These lesions were divided into 5 grades according to the number of lesions and the area of lesions, from "0" to "4". Conclusion: the morphological evaluation of the nervous system requires the pathologists to be familiar with the structure and function of the brain and to master the skills of anatomy and pathology. Therefore, the reliability and accuracy of neurotoxic histopathological evaluation are affected by the scientific and consistent nature of neuropathology and the technical level of operators. The neurovirulence histopathological evaluation method established in this study can be used not only for the non-clinical safety evaluation of the body neurovirulence of the vaccine monkey, but also for the general neurotoxicity test, the animal model of neurodegenerative diseases and the brain. In the evaluation of spinal cord injury animal model, it is a systematic, standard and practical method for evaluating neurotoxicity histopathology.
