【摘要】：目的:制備并鑒定石杉堿甲單克隆抗體,研制石杉堿甲間接競爭酶聯(lián)免疫試劑盒。方法:以石杉堿甲人工抗原免疫BALB/c小鼠獲得單克隆抗體,采用酶聯(lián)免疫吸附法制備石杉堿甲試劑盒,對試劑盒各項指標進行評價。結(jié)果:試劑盒工作范圍為5~2 500 ng/ml,檢測限為6.242 ng/ml,與多種生物堿均無交叉反應,試劑盒可在4℃或-20℃下穩(wěn)定貯存半年以上,石杉堿甲片回收率在94.2%~108.6%之間,變異系數(shù)在2.3%~4.8%之間。結(jié)論:石杉堿甲ELISA試劑盒快速、靈敏、穩(wěn)定,可用于石杉堿甲的含量測定。
[Abstract]:Aim: to prepare and identify monoclonal antibody against Huperzine A and to develop an indirect competitive enzyme linked immunosorbent kit (Elisa) for Huperzine A. Methods: monoclonal antibodies were obtained from BALB / c mice immunized with Huperzine A artificial antigen, and Huperzine A kit was prepared by enzyme-linked immunosorbent assay (Elisa).
