【摘要】：目的:觀察大鼠骨髓間充質(zhì)干細(xì)胞向成骨細(xì)胞誘導(dǎo)分化的效果。方法:采用全骨髓貼壁法培養(yǎng)SD大鼠骨髓間充質(zhì)干細(xì)胞,把生長(zhǎng)狀態(tài)良好的第3代骨髓間充質(zhì)干細(xì)胞,分為兩組,對(duì)照組用DMEM-LG培養(yǎng)基培養(yǎng);實(shí)驗(yàn)組用含有成骨誘導(dǎo)劑的DMEM-LG培養(yǎng)基培養(yǎng)。結(jié)果:全骨髓貼壁法培養(yǎng)的原代骨髓間充質(zhì)干細(xì)胞貼壁且呈梭形;經(jīng)成骨誘導(dǎo)劑誘導(dǎo)后骨髓間充質(zhì)干細(xì)胞呈卵圓形貼壁生長(zhǎng),堿性磷酸酶活性明顯強(qiáng)于對(duì)照組(P0.01),茜素紅染色出現(xiàn)陽(yáng)性的鈣化結(jié)節(jié)。結(jié)論:全骨髓貼壁法培養(yǎng)骨髓間充質(zhì)干細(xì)胞方法簡(jiǎn)單、實(shí)用,所培養(yǎng)的骨髓間充質(zhì)干細(xì)胞在成骨誘導(dǎo)后表現(xiàn)了成骨細(xì)胞的形態(tài)學(xué)和生物學(xué)特性。
[Abstract]:Aim: to observe the differentiation of rat bone marrow mesenchymal stem cells into osteoblasts. Methods: bone marrow mesenchymal stem cells of SD rats were cultured by whole bone marrow adherent method. The third generation of bone marrow mesenchymal stem cells were divided into two groups. The control group was cultured on DMEM-LG medium. The experimental group was cultured on DMEM-LG medium containing osteogenic inducer. Results: primary bone marrow mesenchymal stem cells cultured by whole bone marrow adherent method were fusiform and oval adherent to bone marrow mesenchymal stem cells induced by osteogenic inducer. Alkaline phosphatase activity was significantly higher than that of control group (P0.01). Alizarin red staining showed calcified nodules. Conclusion: bone marrow mesenchymal stem cells cultured by whole bone marrow adherent method is simple and practical. The cultured bone marrow mesenchymal stem cells exhibit the morphological and biological characteristics of osteoblasts after osteogenesis induction.
