【摘要】: 目的研究中樞和外周的促胰泌素(secretin)對(duì)孤束核中胃腸相關(guān)神經(jīng)元活動(dòng)的影響及其機(jī)制。 方法1.記錄烏拉坦麻醉的大鼠孤束核中與電刺激膈下迷走神經(jīng)相關(guān)的神經(jīng)元自發(fā)和誘發(fā)放電。觀察在外周靜脈分別注射不同劑量的secretin(0.04、0.08、0.16μg·kg-1),了解外周外源性secretin對(duì)孤束核中與電刺激膈下迷走神經(jīng)相關(guān)神經(jīng)元的放電的作用。2.記錄烏拉坦麻醉的大鼠孤束核中與擴(kuò)胃相關(guān)的神經(jīng)元的自發(fā)和誘發(fā)放電。觀察在外周靜脈分別注射不同劑量的secretin(0.04、0.08、0.16μg·kg-1)和兔促胰液素抗血清(secretin antiserum)對(duì)該類神經(jīng)元放電的影響。 3.記錄烏拉坦麻醉的大鼠孤束核中與擴(kuò)胃相關(guān)的神經(jīng)元的自發(fā)和誘發(fā)放電,觀察在側(cè)腦室注射微量secretin(0.1μg·ml secretin 1μl)后對(duì)該類細(xì)胞自發(fā)和誘發(fā)放電的影響。4.記錄烏拉坦麻醉的大鼠孤束核中與電刺激膈下迷走神經(jīng)相關(guān)的神經(jīng)元自發(fā)和誘發(fā)放電,觀察在側(cè)腦室注射微量secretin(0.1μg·ml secretin 1μl)后對(duì)該類細(xì)胞自發(fā)和誘發(fā)放電的影響。5.用高效液相色譜法測(cè)定腦脊液中的谷氨酸(Glu)和γ-氨基丁酸(GABA)的含量,觀察在靜脈給予一定劑量secretin對(duì)腦脊液中Glu和GABA的含量的影響,以及給予不同劑量的secretin對(duì)反復(fù)擴(kuò)胃后腦脊液中Glu和
[Abstract]:Aim to study the effects of central and peripheral secretin on gastrointestinal related neurons in nucleus tractus solitarii and its mechanism. Method 1. Spontaneous and evoked discharges of neurons associated with electrical stimulation of the subphrenic vagus nerve were recorded in the nucleus tractus solitarii of rats anesthetized with uratan. To observe the effect of exogenous secretin on the discharge of subphrenic vagus nerve related neurons in nucleus of solitary tract by injecting different doses of secretinus 0.04 (0.08 渭 g 路kg ~ (-1) into peripheral vein respectively, and to investigate the effect of exogenous secretin on the discharges of subphrenic vagus neurons in the nucleus tractus solitarii. Spontaneous and evoked discharges of neurons associated with gastric dilatation in the nucleus tractus solitarius were recorded. The effects of different doses of secretinine 0.04 (0.08 渭 g 路kg ~ (-1) and rabbit antiserum secretin antiserum3 on the discharge of these neurons were observed. The spontaneous and evoked discharges of neurons associated with gastric distending in the nucleus tractus solitarius were recorded and the effects of intraventricular injection of secretin(0.1 渭 g ml secretin 1 渭 l on the spontaneous and evoked discharges of these neurons were observed. The spontaneous and evoked discharges of neurons associated with electrical stimulation of subphrenic vagus nerve were recorded in the nucleus tractus solitarii of rats anesthetized by urartan. The effects of microinjection of secretin(0.1 渭 g ml secretin 1 渭 l into lateral ventricle on the spontaneous and evoked discharges of these neurons were observed. The content of glutamate Gluand 緯 -aminobutyrate in cerebrospinal fluid (CSF) was determined by high performance liquid chromatography (HPLC). The effects of intravenous administration of secretin on the contents of Glu and GABA in cerebrospinal fluid (CSF) were observed.
