【摘要】: 本研究旨在闡明;蛆Z去氧膽酸(TCDCA)對小鼠免疫細胞凋亡的影響及其影響機理。采用二苯胺法檢測了給藥后小鼠不同免疫細胞的凋亡百分率,結(jié)果顯示:低劑量TCDCA能夠顯著抑制正常小鼠腹腔巨噬細胞、脾臟淋巴細胞和血液淋巴細胞的凋亡,但對胸腺細胞凋亡無顯著影響;高劑量TCDCA能夠顯著抑制正常小鼠外周血淋巴細胞的凋亡,對脾臟淋巴細胞凋亡無顯著影響,但能夠顯著促進正常小鼠腹腔巨噬細胞和胸腺細胞的凋亡。表明TCDCA對不同的免疫細胞的凋亡具有不同的影響,顯示出其免疫調(diào)節(jié)作用。 通過考察TCDCA對外周血淋巴細胞凋亡過程中Caspase依賴的內(nèi)源性細胞凋亡途徑的影響,探討了其影響小鼠免疫細胞凋亡的機理。采用流式細胞儀檢測了TCDCA對外周血液淋巴細胞凋亡早期磷脂酰絲氨酸外翻百分率的影響,證明低劑量和高劑量TCDCA均能夠降低磷脂酰絲氨酸外翻百分率,用藥后第4天,TCDCA對磷脂酰絲氨酸外翻百分率的抑制作用最顯著;采用流式細胞儀檢測了TCDCA對外周血淋巴細胞線粒體膜電位發(fā)生變化的百分率,證明低劑量TCDCA能夠極顯著的降低淋巴細胞線粒體膜勢能變化百分率;采用分光光度法檢測了TCDCA對外周血淋巴細胞凋亡過程中Caspase—3、Caspase—9蛋白酶的活化程度的影響,證明高、低劑量TCDCA均能極顯著的抑制Caspase-9及Caspase-3蛋白酶活性。實驗結(jié)果表明TCDCA對小鼠外周血淋巴細胞Caspase依賴的內(nèi)源性細胞凋亡途徑的不同時期、不同階段均有顯著的凋亡抑制作用。
[Abstract]:The aim of this study was to elucidate the effect of TCDCA on the apoptosis of immune cells in mice and the mechanism of its influence. The percentage of apoptosis in different immune cells of mice was detected by two aniline method. The results showed that the low dose TCDCA could significantly inhibit the peritoneal macrophages, spleen lymphocytes and blood lymphocytes in normal mice. Apoptosis has no significant effect on the apoptosis of thymus cells. High dose TCDCA can significantly inhibit apoptosis of peripheral blood lymphocytes in normal mice and have no significant influence on the apoptosis of spleen lymphocytes, but can significantly promote apoptosis of peritoneal macrophages and thymocytes in normal mice. It is indicated that TCDCA has no effect on the apoptosis of different immune cells. The same effect shows its immunomodulatory effect.
By investigating the effect of Caspase dependent endogenous apoptosis pathway on the apoptosis of peripheral blood lymphocyte in TCDCA, the mechanism of its influence on the apoptosis of immune cells in mice was investigated. The effect of TCDCA on the percentage of phosphatidyl serine ectropion in the early stage of apoptosis was detected by flow cytometry. The percentage of phosphatidylserine ectropion could be reduced by dose of TCDCA, and the inhibitory effect of TCDCA on the percentage of phosphatidylserine ectropion was the most significant after fourth days of drug use. The percentage of mitochondrial membrane potential changes in peripheral blood lymphocytes of TCDCA was detected by flow cytometry. It was proved that low dose TCDCA could significantly reduce lymphatic thinning. The percentage of mitochondrial membrane potential energy changes; the effect of the activation degree of Caspase - 3, Caspase - 9 protease in the apoptotic process of TCDCA peripheral blood lymphocytes was detected by spectrophotometric method. It was proved that high and low dose TCDCA could significantly inhibit the activity of Caspase-9 and Caspase-3 protease. The experimental results showed that TCDCA was in the peripheral blood of mice. In different stages of Caspase dependent endogenous apoptosis pathway, apoptosis of the Ba cells was significantly inhibited at different stages.
