【摘要】:目的 探討胰腺發(fā)育中胰島細(xì)胞的分化、遷移和胰島的形成,以及在此過(guò)程中相關(guān)基因表達(dá)的變化。對(duì)胰島β細(xì)胞損傷后的胰島再生機(jī)制進(jìn)行初步研究,并檢測(cè)了β細(xì)胞損傷后調(diào)控胚胎胰島細(xì)胞分化發(fā)育的部分關(guān)鍵性基因Pdx-1、Ngn3、Nkx2.2及Nkx6.1的表達(dá)情況,為應(yīng)用胰腺干細(xì)胞作為糖尿病生物治療性手段提供基礎(chǔ)性研究。 材料和方法 1.收集征得孕婦簽字同意后、因意外等各方面原因必須終止妊娠的不同胎齡段的正常胚胎標(biāo)本進(jìn)行組織學(xué)觀察,并通過(guò)免疫組織化學(xué)方法觀察胰島細(xì)胞的分化發(fā)育和CD34、AFP、IGF-I、IGF-IR,TGF-β、TGF-βR,bFGF及FGFR因子在胰腺發(fā)育中的表達(dá)分布。 2.采用β細(xì)胞毒性劑—Alloxan腹腔注射破壞新生SD大鼠的β細(xì)胞,復(fù)制糖尿病模型,采用免疫組織化學(xué)方法觀察胰島β細(xì)胞的再生,并通過(guò)RT-PCR、Western-blot方法檢測(cè)Pdx-1、Ngn3、Nkx2.2及Nkx6.1基因的表達(dá)。 結(jié)果 1.人胚胎觀察 ① 4周時(shí),,人胚胰芽由前腸末端發(fā)出,腹、背胰在胎齡5周時(shí)融
[Abstract]:Objective to investigate the differentiation, migration and formation of pancreatic islets during pancreatic development, and the changes of gene expression in the process. The mechanism of islet regeneration after 尾 -cell injury was studied, and the expression of some key genes Pdx-1, Ngn3Ngn3Nkx2.2 and Nkx6.1, which regulated the differentiation and development of embryonic islet cells after 尾 -cell injury, was detected. To provide basic research for the application of pancreatic stem cells as a biotherapeutic method for diabetes mellitus. Materials and methods 1. To collect and obtain the consent of the pregnant woman, to observe the histology of the normal embryo at different stages of gestational age that must be terminated due to accident and other reasons, The differentiation and development of islet cells and the expression of TGF- 尾 TGF- 尾 TGF- 尾 TGF- 尾 and FGFR factor in pancreatic development were observed by immunohistochemical method. 2. 尾 -Alloxan, a cytotoxic agent, was injected intraperitoneally to destroy the 尾 cells of newborn SD rats, and the diabetic model was established. The regeneration of islet 尾 cells was observed by immunohistochemical method. The expression of Nkx2.2 and Nkx6.1 genes was detected by RT-PCRG-Western-blot. Results 1.Results 1. When human embryo was observed for 14 weeks, the bud of human embryo came from the end of the foregut, and the abdomen and dorsal pancreas melted at 5 weeks of gestational age.
