【摘要】:堿性成纖維細(xì)胞生長(zhǎng)因子和轉(zhuǎn)化生長(zhǎng)因子β1對(duì)體外培養(yǎng)幼兔關(guān)節(jié)軟骨細(xì)胞增殖的影響 目的 堿性成纖維細(xì)胞生長(zhǎng)因子和轉(zhuǎn)化生長(zhǎng)因子β1與軟骨細(xì)胞的生長(zhǎng)、分化十分密切,因此在軟骨組織工程研究中受到了普遍的關(guān)注。本文研究堿性成纖維細(xì)胞生長(zhǎng)因子(bFGF)和轉(zhuǎn)化生長(zhǎng)因子β1(TGF-β1)在單一和聯(lián)合應(yīng)用時(shí),對(duì)體外培養(yǎng)幼兔關(guān)節(jié)軟骨細(xì)胞增殖的影響,探討軟骨組織工程中能夠有效促進(jìn)軟骨細(xì)胞增殖的兩種生長(zhǎng)因子的濃度組合方式。 方法 體外多代培養(yǎng)的幼兔關(guān)節(jié)軟骨細(xì)胞(第2、4、6、8代)分別以bFGF(1,5,10ng/ml)、TGF-β1(0.1,0.5,1ng/ml)及其不同濃度間兩兩組合的聯(lián)合刺激組為實(shí)驗(yàn)組,以常規(guī)培養(yǎng)組為對(duì)照組,通過(guò)CellTiter96(?)AQueous單溶液細(xì)胞增殖分析法測(cè)定各代細(xì)胞在生長(zhǎng)因子(及其聯(lián)合)作用下的增殖效應(yīng)。 結(jié)果 單獨(dú)應(yīng)用時(shí),1ng/ml的TGF-β1為促增殖的最佳效應(yīng)濃度;bFGF在第2、4、6代細(xì)胞中的促增殖最佳濃度為10ng/ml,在第8代細(xì)胞中的最佳濃度為5ng/ml;聯(lián)合應(yīng)用時(shí),當(dāng)TGF-β1為低濃度(0.1ng/ml)或中濃度(0.5ng/ml)聯(lián)合bFGF時(shí),高濃度bFGF(10ng/ml)與之組合促增殖效果最佳;當(dāng)高濃度TGF-β1(1ng/ml)與bFGF聯(lián)合時(shí),對(duì)早期(第2、4代)細(xì)胞,TGF-β1(1ng/ml)+bFGF(10ng/ml)為最佳的促增殖濃度組合,而在后期(第6、8代),,TGF-β1(1ng/ml)
[Abstract]:Effects of basic fibroblast growth factor and transforming growth factor beta 1 on the proliferation of rabbit articular chondrocytes in vitro
Objective basic fibroblast growth factor (GGF) and transforming growth factor beta 1 (TGF - beta 1) are closely related to the growth and differentiation of chondrocytes. Therefore, it is widely concerned in the study of cartilage tissue engineering. In this paper, basic fibroblast growth factor (bFGF) and transforming growth factor beta 1 (TGF- beta 1) were studied in a single and combined application to the culture in vitro. The effects of two kinds of growth factors on chondrocyte proliferation in cartilage tissue engineering were discussed.
Methods the chondrocytes of young rabbit joint in vitro (2,4,6,8) were treated with bFGF (1,5,10ng / ml), TGF- beta 1 (0.1,0.5,1ng / ml) and the combination of 22 combinations of different concentrations as the experimental group. The normal culture group was used as the control group. The cells of each generation were determined by the CellTiter96 (?) AQueous single solution cell proliferation analysis. Proliferation effect under the action of long factor (and its combined).
Results the optimal concentration of 1ng / ml TGF- beta 1 was the best effect concentration for promoting proliferation, and the best concentration of bFGF in 2,4,6 cells was 10NG / ml, and the best concentration in the eighth generation cells was 5ng / ml. When the TGF- beta 1 was low concentration (0.1ng / ml) or medium concentration L) is the best combination to promote proliferation. When the high concentration of TGF- beta 1 (1ng / ml) is combined with bFGF, the optimal proliferation concentration combination for the early (2,4) cells, TGF- beta 1 (1ng / ml) +bFGF (10NG / ml), and in the later period (1).
