實驗動物墊料的微生物污染監(jiān)測及影響因素研究

【摘要】:目的對實驗動物墊料進行微生物指標監(jiān)測,為管理機構制定墊料的衛(wèi)生標準提供依據,為墊料生產企業(yè)在加工、滅菌、包裝、運輸、儲存等環(huán)節(jié)及動物使用單位對動物實驗結果影響因素分析方面提供參考依據。方法對來自不同實驗動物生產和使用單位的墊料,參照GB4789-2010、2012檢測方法進行菌落總數(shù)、霉菌及大腸桿菌計數(shù)、沙門菌的檢測。結果 49份未滅菌的墊料存在不同程度的細菌、霉菌及大腸桿菌污染,未檢出沙門菌;不同材質的墊料在不同條件下滅菌,在121℃30 min的條件下具有最佳滅菌效果,各項微生物指標均合格;已滅菌的21份墊料有19份存在不同程度的細菌及霉菌污染,未檢出沙門菌及大腸桿菌;松軟的混合型刨花墊料在滅菌后7 d檢測到細菌;采用敞開編織袋儲存5 d即檢測到細菌污染;墊料使用1 d后的菌量可達104CFU/g,細菌及霉菌含菌量按天數(shù)逐步遞增,但未檢出沙門菌。結論墊料使用前必須消毒滅菌,采用合理的方式儲存,及時更換可改善飼養(yǎng)環(huán)境,可減少微生物污染,保障實驗動物的質量及實驗結果的可靠。
[Abstract]:Objective to monitor the microbiological indexes of the gasket of laboratory animals, and to provide the basis for the establishment of sanitary standard of the gasket by the management organization, and to provide the basis for the processing, sterilization, packing and transportation of the gasket material production enterprise. Storage and animal use units provide a reference for the analysis of the factors affecting the results of animal experiments. Methods according to GB4789-20102012, the total number of bacteria, the count of mold and Escherichia coli, and the salmonella were detected from different laboratory animal production and use units. Results 49 unsterilized mats were contaminated by bacteria, mold and Escherichia coli, and salmonella was not detected. The best sterilizing effect was obtained by sterilizing the mats of different materials under different conditions at 121 鈩,
