新型CpG ODN的抗病毒作用及其作為病毒疫苗佐劑的實驗研究

【摘要】: 含有非甲基化CpG基序的寡聚脫氧核苷酸(CpG ODN),能刺激哺乳動物免疫細(xì)胞,誘導(dǎo)Th1途徑的免疫應(yīng)答。A型CpG ODN能刺激人漿細(xì)胞樣樹突狀細(xì)胞產(chǎn)生大量IFN-α,強(qiáng)烈激活NK細(xì)胞;B型CpG ODN能刺激B細(xì)胞增殖,分泌細(xì)胞因子;C型CpG ODN兼有A型和B型的作用。CpG ODN被廣泛用于治療感染性疾病、癌癥、過敏性疾病,以及疫苗佐劑的研究,并已取得了肯定的結(jié)果。 我們對本室設(shè)計的C型CpG ODN BW001誘導(dǎo)產(chǎn)生抗病毒作用的機(jī)理進(jìn)行了探討,BW001能刺激人PBMC表達(dá)IFN-β、IFN-ω、IFN-γ以及至少10種IFN-α亞型的mRNA。進(jìn)一步研究發(fā)現(xiàn),BW001能誘生抗柯薩奇病毒B3的作用。我們還對本室設(shè)計的不同類型CpG ODN作為乙肝疫苗佐劑的作用進(jìn)行了研究,結(jié)果A型、B型和C型CpG ODN均能顯著促進(jìn)特異性抗體的產(chǎn)生,且IgG類型以IgG2a為主,刺激了Th1途徑的免疫應(yīng)答。上述研究為研制我國自主知識產(chǎn)權(quán)的防治病毒感染性疾病的新藥提供了實驗基礎(chǔ)。
[Abstract]:Oligodeoxynucleotides containing unmethylated CpG motifs, CpG ODNs, stimulate mammalian immune cells. The immune response induced by Th1 pathway. Type A CpG ODN can stimulate the production of a large number of IFN- 偽 in human plasma cell-like dendritic cells, and strongly activate NK cells, type B CpG ODN can stimulate the proliferation of B cells. Secretory cytokine type C CpG ODN has both type A and B functions. CpG ODN has been widely used in the treatment of infectious diseases, cancer, allergic diseases, and vaccine adjuvants. The mechanism of induction of antiviral effects by type C CpG ODN BW001 designed in our laboratory was studied. The mechanism of BW001 stimulating the expression of IFN- 尾 -IFN- 蠅 IFN- 緯 and at least 10 kinds of mRN- 偽 subtypes in human PBMC was investigated. Further studies showed that BW001 could induce resistance to Coxsackie virus B 3. We also studied the effects of different types of CpG ODN as adjuvant of hepatitis B vaccine in our laboratory. The results showed that type A, B and C CpG ODN could significantly promote the production of specific antibodies, and IgG2a was the main type of IgG. It stimulates the immune response of Th1 pathway. These studies provide experimental basis for the development of new drugs for the prevention and treatment of viral infectious diseases with independent intellectual property rights in China.
