【摘要】: 人類子宮內(nèi)膜對(duì)胚胎的接受性存在時(shí)間和空間的短暫性,即胚胎著床活動(dòng)受到“種植窗”的限制,“種植窗”的啟動(dòng)常常發(fā)生在內(nèi)源性黃體生成素(luteinizing hormone, LH)峰或使用外源性人絨毛促性腺激素(human chorionic gonadotropin, hCG)(LH的類似物)或黃體酮的第4-5天,關(guān)閉發(fā)生在第9-10天。進(jìn)入到宮腔的胚胎和子宮內(nèi)膜按一定的時(shí)、空順序分泌相關(guān)蛋白或/和局部因子從而達(dá)到相互識(shí)別、相互融合完成著床過(guò)程。胚胎著床過(guò)程的限速步驟是胚胎發(fā)育與子宮內(nèi)膜向接受態(tài)轉(zhuǎn)化過(guò)程同步。而LH峰是促進(jìn)卵子成熟、胚胎發(fā)育和子宮內(nèi)膜向接受態(tài)同步轉(zhuǎn)化的、最關(guān)鍵的調(diào)節(jié)因素。 如何評(píng)估胚胎發(fā)育與子宮內(nèi)膜向接受態(tài)同步轉(zhuǎn)化是預(yù)測(cè)胚胎著床成功與否的重要指標(biāo)。 現(xiàn)行評(píng)估胚胎發(fā)育潛能的方法包括胚胎形態(tài)學(xué)評(píng)分和生化指標(biāo)檢測(cè)。然而胚胎評(píng)分與胚胎發(fā)育潛能并不完全正相關(guān);胚胎分泌的細(xì)胞因子和蛋白類物質(zhì)參與自身發(fā)育調(diào)節(jié)和胚胎著床過(guò)程,被預(yù)測(cè)可能用來(lái)衡量胚胎發(fā)育潛能的標(biāo)志物。本論文將在我們前期工作基礎(chǔ)上進(jìn)一步進(jìn)行人類卵母細(xì)胞和種植前胚胎LH表達(dá)的研究,為探討LH在人類卵子和胚胎發(fā)育過(guò)程中的調(diào)節(jié)作用提供實(shí)驗(yàn)依據(jù)。上個(gè)世紀(jì)五十年代開(kāi)始,學(xué)者們就從組織形態(tài)學(xué)、亞細(xì)胞結(jié)構(gòu)、局部因子表達(dá)及其類固醇激素的調(diào)節(jié)作用等層次對(duì)子宮內(nèi)膜胚胎接受性的評(píng)價(jià)方面做了大量研究,提出上皮細(xì)胞吞飲泡(pinopod)、整合素αυβ3、白血病抑制因子(LIF)、血管內(nèi)皮生長(zhǎng)因子(VEGF)的表達(dá)部分地反映子宮內(nèi)膜對(duì)胚胎的接受性。但由
[Abstract]:The temporal and spatial transience of human endometrial receptivity to embryos, that is, the implantation of embryos is limited by the "implantation window". The initiation of "implant window" often occurs on the 4-5 day of endogenous luteinizing hormone (LHK) peak or the use of exogenous human chorionic gonadotropin (hCG)(LH analogue) or progesterone, and the closure occurs on the 9th to 10th day. When the embryo and endometrium enter the uterine cavity according to a certain order the empty sequence secretes the related protein or / and local factor to achieve mutual recognition and fusion to complete the implantation process. The limiting step of embryo implantation is the synchronization of embryo development and endometrial to receptive transformation. LH peak is the most important regulatory factor to promote the ovum maturation, embryo development and endometrial synchronous transformation to the receptive state. How to evaluate the synchronous transformation between embryo development and endometrium is an important index to predict the success of embryo implantation. Current methods for evaluating embryonic development potential include embryo morphological scores and biochemical indicators. However, embryo score is not positively correlated with embryonic development potential. Cytokines and proteins secreted by embryos are involved in the process of self-regulation and implantation of embryos, which may be used as markers to measure the developmental potential of embryos. This thesis will further study the expression of LH in human oocytes and preimplantation embryos on the basis of our previous work in order to provide experimental basis for exploring the regulatory role of LH in the development of human eggs and embryos. Since the 1950s, researchers have done a great deal of research on the evaluation of endometrial embryo acceptability from the aspects of histomorphology, subcellular structure, local factor expression and the regulation of steroid hormones.
