【摘要】:目的:觀察腺苷三磷酸(ATP)對(duì)大鼠離體胃平滑肌運(yùn)動(dòng)的調(diào)節(jié)作用;觀察ATP對(duì)大鼠離體近端結(jié)腸縱行肌運(yùn)動(dòng)的影響并探討其受體機(jī)制;分析ATP介導(dǎo)大鼠胃體環(huán)行肌收縮反應(yīng)的受體機(jī)制。 方法:制備大鼠離體胃體縱行肌、胃體環(huán)行肌、胃竇縱行肌、胃竇環(huán)行肌和近端結(jié)腸縱行肌標(biāo)本,觀察ATP的作用;觀察觀察ATP、α,β-MeATP和腺苷對(duì)大鼠胃體環(huán)行肌的作用并分析相關(guān)的受體。 結(jié)果:在胃體縱行肌標(biāo)本,ATP(0.1 μmol·L~(-1) -1mmol·L~(-1))誘發(fā)微弱的舒張反應(yīng),繼而出現(xiàn)收縮反應(yīng);在高鉀預(yù)收縮條件下,ATP則引起較大舒張反應(yīng)后續(xù)較小的收縮反應(yīng)。ATP(0.1 μmol·L~(-1)-1 mmol·L~(-1))對(duì)胃體環(huán)行肌誘發(fā)單相收縮反應(yīng)。低濃度ATP(1 μmol·L~(-1) -100 μmol·L~(-1))抑制胃竇縱行肌的自發(fā)性收縮幅度,同時(shí)加快收縮頻率;高濃度ATP(1 mmol·L~(-1))則完全抑制自發(fā)性收縮反應(yīng);在高鉀預(yù)收縮條件下,ATP產(chǎn)生濃度依賴性舒張反應(yīng)。低濃度ATP(1 μmol·L~(-1)-10 μmol·L~(-1))先增大后減低胃竇環(huán)行肌的自發(fā)性收縮幅度,同時(shí)加快收縮頻率;高濃度ATP(100 μmol·L~(-1)-1 mmol·L~(-1))則抑制自發(fā)性收縮反應(yīng)。 在靜息張力時(shí),ATP(0.1 μmol·L~(-1)-1 mmol·L~(-1))誘
[Abstract]:Aim: to observe the effects of adenosine triphosphate (ATP) on the motility of isolated rat gastric smooth muscle, to observe the effect of ATP on the motility of the longitudinal muscle of rat proximal colon and to explore the mechanism of its receptor. Objective: to investigate the receptor mechanism of ATP mediated gastric circular muscle contraction in rats. Methods: the specimens of isolated longitudinal muscle of gastric body, circular muscle of gastric body, longitudinal muscle of gastric antrum, circular muscle of gastric antrum and longitudinal muscle of proximal colon were prepared and the effect of ATP was observed. The effects of ATP, 偽, 尾 -MeATP and adenosine on the circular muscle of gastric body in rats were observed and the related receptors were analyzed. Results: in the longitudinal muscle of gastric body, ATP 0.1 渭 mol / L ~ (-1) -1mmol / L ~ (-1) induced a weak diastolic response and then a contractile response. Under the condition of high potassium precontraction, ATP induced a larger diastolic response, followed by a smaller contractile response. ATP-0.1 渭 mol / L ~ (-1) / L ~ (-1) mmol / L ~ (-1) induced monophasic contraction of the circular muscle of the gastric body. Low concentration of ATP(1 渭 mol L ~ (1) -100 渭 mol / L ~ (-1) inhibited the spontaneous contraction of the longitudinal muscle of gastric antrum and accelerated the contractile frequency, while the high concentration of ATP(1 mmol was completely inhibited the spontaneous contraction, and the concentration of ATP produced a concentration-dependent relaxation under the condition of high potassium precontraction. Low concentration of ATP(1 渭 mol L ~ (1) -10 渭 mol 路L ~ (-1) increased at first, then decreased the amplitude of spontaneous contraction of the antral circular muscle and increased the contractile frequency at the same time, while the high concentration of ATP(100 渭 mol L ~ (1) ~ (-1) mmol L _ (1) inhibited the spontaneous contraction.
