【摘要】：目的觀察病理穿刺活檢組織常規(guī)石蠟和常規(guī)冰凍制片HE染色效果的異同,提高穿刺活檢組織的制片質(zhì)量。方法取中山大學附屬第一醫(yī)院各手術(shù)科送檢的病理穿刺活檢組織標本(骨和骨髓、乳腺、脂肪)進行處理,每份送檢標本均一分為二,一份常規(guī)固定脫水包埋切片;一份常規(guī)冰凍切片,兩份標本切片均常規(guī)HE染色,鏡下觀察閱片并作比較。結(jié)果多數(shù)穿刺活檢組織常規(guī)石蠟和常規(guī)冰凍切片制成的HE片結(jié)構(gòu)完整,厚薄均勻,染色恰當,形態(tài)清晰,無斷裂破碎現(xiàn)象,核漿分明,切片平整,定位準確。骨和骨髓穿刺活檢組織不宜進行常規(guī)冰凍切片,也不能直接進行常規(guī)石蠟切片,必須經(jīng)過脫鈣液脫鈣才能切片;含脂肪多的乳腺穿刺活檢組織和脂肪組織做常規(guī)石蠟切片,脫水前必須用純丙酮脫脂才能切片;而進行常規(guī)冰凍切片,組織切片不完整、折疊、破碎、脫落,嚴重影響制片質(zhì)量和診斷的正確性。結(jié)論大多數(shù)病理穿刺活檢組織都可以進行常規(guī)石蠟和常規(guī)冰凍切片,切片HE染色質(zhì)量良好。
[Abstract]:Objective to observe the difference of HE staining effect between routine paraffin and frozen biopsy tissue and to improve the quality of biopsy tissue. Methods the biopsy specimens (bone and bone marrow, mammary gland, fat) of the first affiliated Hospital of Sun Yat-sen University were taken for treatment. Each specimen was divided into two parts, and a routine fixed dehydration embedded section was made. One routine frozen section and two specimens were stained with HE. Results the HE slices made from routine paraffin and frozen sections of most biopsy tissues were of complete structure, uniform thickness, proper staining, clear morphology, no fracture and fragmentation, clear nucleus and cytoplasm, flat sections and accurate orientation. Bone and bone marrow biopsy tissues are not suitable for routine frozen sections, nor can conventional paraffin sections be carried out directly. Decalcification of bone and bone marrow biopsy tissues must be done through decalcification solution before they can be sliced. Before dehydration, it is necessary to use pure acetone to defatted the sections, while the conventional frozen sections are not complete, folded, broken and shedded, which seriously affect the quality of film preparation and the correctness of diagnosis. Conclusion conventional paraffin and frozen sections can be performed in most pathological biopsy tissues, and the HE staining quality of the sections is good.
