【摘要】:流感病毒是最常見的呼吸道致病因子,每次流感大流行的時(shí)候都有很多人死亡,例如在1918年的流感大流行就有2000萬(wàn)人死于流感。流感病毒常見的宿主有人、豬、馬和禽類,并可以在不同宿主之間交叉?zhèn)鞑?如1997年香港的流感病毒就是由雞傳播給人的,在18個(gè)感染流感的人中就有6個(gè)死亡。疫苗是預(yù)防流感的一種好方法,目前最常用的是滅活疫苗(Ivac),其次是減毒活疫苗(Livac)和亞單位疫苗(Subvac)。流感DNA疫苗是一種新型的預(yù)防流感的疫苗,目前正在很多國(guó)家的實(shí)驗(yàn)室中進(jìn)行研究,部分已經(jīng)在動(dòng)物實(shí)驗(yàn)中獲得了良好效果。 DNA疫苗能夠同時(shí)刺激機(jī)體B細(xì)胞,輔助性T細(xì)胞和細(xì)胞毒性T細(xì)胞,產(chǎn)生特異性體液免疫和細(xì)胞免疫,因而受到大家關(guān)注。美國(guó)國(guó)立衛(wèi)生研究院(NIH)和世界衛(wèi)生組織(WHO)都強(qiáng)調(diào)流感DNA疫苗是流感疫苗研究的一個(gè)重要方向,流感病毒血凝素(hemagglutinin,HA)基因的研究正是該重要研究?jī)?nèi)容之一。流感病毒基因組第4節(jié)段編碼HA,人和動(dòng)物流感病毒的HA刺激機(jī)體產(chǎn)生最主要的保護(hù)性抗體,此抗體具有中和病毒和阻止病毒穿入靶細(xì)胞的作用,能中斷病毒的復(fù)制。HA分為重鏈HA1和輕鏈HA2區(qū),HA1區(qū)是該蛋白與宿主細(xì)胞受體結(jié)合部分,HA2區(qū)是具有膜融合活性的部分。本文對(duì)流感病毒HA1區(qū)進(jìn)
[Abstract]:Influenza viruses are the most common respiratory factor, with many people dying during each pandemic, such as the 1918 pandemic, which killed 20 million people. Influenza virus is a common host of people, pigs, horses and birds, and can be spread across different hosts. For example, in 1997, the influenza virus in Hong Kong was transmitted from chickens to humans, and six of the 18 people infected with the flu died. Vaccine is a good way to prevent influenza. At present, the most commonly used vaccine is inactivated vaccine Ivaca, followed by live attenuated vaccine (livaca) and subunit vaccine. Influenza DNA vaccine is a new vaccine to prevent influenza. It is being studied in many laboratories in many countries, and some of it has achieved good results in animal experiments. DNA vaccine can stimulate B cells, helper T cells and cytotoxic T cells at the same time and produce specific humoral and cellular immunity. Both the National Institutes of Health (NIH) and the World Health Organization (WHO) have emphasized that influenza DNA vaccine is an important direction of influenza vaccine research, and the study of influenza virus hemagglutinin (HA) gene is one of the important research contents. Section 4 of the influenza virus genome encodes HA.The HA of human and animal influenza viruses stimulates the body to produce the most important protective antibody, which neutralizes the virus and prevents it from penetrating into the target cells.
