【摘要】: 目的通過一些體內(nèi)和體外試驗證實,來自細(xì)菌或人工合成的非甲基化的CpG寡核苷酸(ODN)能誘導(dǎo)免疫淋巴細(xì)胞增殖及免疫球蛋白、單核細(xì)胞因子、INF-γ等的分泌,并能激活自然殺傷細(xì)胞的溶細(xì)胞效應(yīng)。CpG ODN的這些功能意味它可能在增強機體對疫苗的免疫應(yīng)答方面具有一定的前景,F(xiàn)有的HBsAg重組蛋白疫苗雖說在一定程度上能起到較好的作用,但一些調(diào)查統(tǒng)計表明,在經(jīng)過此種疫苗接種的人群中,約有10%~15%的免疫人群并未產(chǎn)生較理想的免疫效果。正因為如此,許多研究人員正試圖從新型免疫佐劑的開發(fā)這一方向?qū)ふ铱朔@個缺點的新型疫苗。 本實驗就是圍繞這一主題試圖通過CpG寡核苷酸(CpG ODN)與乙型肝炎病毒重組蛋白疫苗聯(lián)合免疫C57BL/6小鼠,觀察它對小鼠免疫狀況的影響。進而探討CpG寡核苷酸(CpG ODN)能否作為一種佐劑與HBsAg疫苗一起,更好地刺激機體免疫系統(tǒng)產(chǎn)生有效的應(yīng)答。 方法將小鼠分為三組,第一組為生理鹽水對照,第二組為乙型肝炎病毒重組蛋白疫苗接種,第三組為CpG ODN與重組HBsAg疫苗聯(lián)合接種,按照第1天、第15天、第22天的程序進行免疫接種,于免疫后第29天分別采血1.0ml,并處死小鼠迅速制備脾細(xì)胞懸液。用MTT法分別進行HBsAg蛋白特異性刺激的淋巴細(xì)胞轉(zhuǎn)化試驗、HBsAg蛋白特異性刺激的細(xì)胞毒性T細(xì)胞殺傷試驗(HBsAg specific
[Abstract]:Objective to study in vitro and in vivo studies that CpG oligodeoxynucleotides (ODNs) from bacteria or synthetic unmethylated CpG can induce the proliferation of immune lymphocytes and the secretion of immunoglobulin, monocyte cytokines, such as INF- 緯, etc. These functions of CpG ODN, which can activate the cytotoxic effect of natural killer cells, may have a certain prospect in enhancing the immune response to the vaccine. Although the existing HBsAg recombinant protein vaccine can play a better role to some extent, some surveys and statistics show that about 10% or 15% of the population vaccinated with this vaccine did not have an ideal immune effect. Because of this, many researchers are trying to find new vaccines to overcome this shortcoming in the direction of developing new immune adjuvants. The purpose of this study was to immunize C57BL/6 mice with recombinant protein vaccine of hepatitis B virus by CpG oligodeoxynucleotide (CpG ODN) and to observe its effect on the immune status of mice. To explore whether CpG oligonucleotide can be used as an adjuvant with HBsAg vaccine to stimulate the immune system to produce an effective response. Methods mice were divided into three groups, the first group was saline control group, the second group was hepatitis B virus recombinant protein vaccine, the third group was CpG ODN combined with recombinant HBsAg vaccine, according to the 1st day, 15th day. On the 29th day after immunization, 1.0 ml blood was collected, and mice were killed to prepare spleen cell suspension.
