抗人骨髓間充質(zhì)干細(xì)胞單克隆抗體ZUC3在大鼠骨髓間充質(zhì)干細(xì)胞生物學(xué)特性研究中的應(yīng)用

【摘要】:間充質(zhì)干細(xì)胞(mesenchymal stem cells,MSCs)因其自我更新、多向分化能力和低免疫原性,成為組織工程、再生醫(yī)學(xué)的理想種子細(xì)胞,并在越來越多的臨床疾病治療中發(fā)揮作用。目前除人MSCs外,許多其他種屬來源的MSCs也已有實(shí)驗(yàn)報(bào)道,如貓、狗、狒狒、兔、豬、山羊、綿羊等,但在動(dòng)物模型研究中最常用的是大鼠。 作為MSCs鑒定的重要組成部分,其特異性的表面標(biāo)記一直以來都是人們研究的熱點(diǎn),但至今尚未有何突破性的進(jìn)展。通過單克隆抗體的篩選制備來尋找特異性表面分子是近年來開展的前沿技術(shù)。本課題小組通過該方法以人骨髓MSCs免疫小鼠,通過雜交瘤技術(shù)獲得了穩(wěn)定分泌抗人骨髓MSCs抗體的細(xì)胞株5株,分別將其分泌的抗體命名為ZUB1、ZUB4、ZUC3、ZUE12和ZUF10。在對(duì)5種抗體進(jìn)行的特異性檢測(cè)中發(fā)現(xiàn),它們與除骨髓外的人體其他組織以及人骨髓中的其他細(xì)胞成分均不存在交叉反應(yīng),但ZUC3與大鼠和犬的骨髓MSCs存在明顯的交叉反應(yīng),不具有種屬特異性。 本研究旨在建立大鼠MSCs的體外培養(yǎng)與鑒定方法,分析ZUC3在大鼠MSCs檢測(cè)鑒定中的價(jià)值,并應(yīng)用ZUC3對(duì)處于不同生長(zhǎng)階段及向不同方向分化的大鼠MSCs作進(jìn)一步檢測(cè),以了解ZUC3相應(yīng)抗原在大鼠MSCs中的表達(dá)規(guī)律。研究分兩部分:一、大鼠骨髓間充質(zhì)干細(xì)胞的體外培養(yǎng)擴(kuò)增和生物學(xué)特性研究,二、
[Abstract]:Mesenchymal stem cells (MSCs) have become ideal seed cells for tissue engineering and regenerative medicine because of their self-renewal, multi-directional differentiation and low immunogenicity, and play an important role in the treatment of more and more clinical diseases. At present, in addition to human MSCs, many other species of MSCs have been reported, such as cats, dogs, baboons, rabbits, pigs, goats, sheep and so on. As an important part of MSCs identification, its specific surface marker has been a hot research topic, but no breakthrough has been made so far. Screening and preparation of monoclonal antibodies to find specific surface molecules is a frontier technology developed in recent years. In this study, we immunized mice with human bone marrow MSCs and obtained 5 cell lines which secreted anti-human bone marrow MSCs antibody stably by hybridoma technique, and named ZUB1ZUB4ZUC3UC3UU12 and ZUF10respectively. In the specific detection of five antibodies, it was found that they did not cross react with other tissues except bone marrow and other cell components in human bone marrow, but ZUC3 had obvious cross reaction with rat and dog bone marrow MSCs. It has no species specificity. The purpose of this study was to establish an in vitro culture and identification method for rat MSCs, to analyze the value of ZUC3 in the detection and identification of rat MSCs, and to further detect rat MSCs in different growth stages and in different directions by ZUC3. To understand the expression of ZUC3 antigens in rat MSCs. The study is divided into two parts: first, the proliferation and biological characteristics of rat bone marrow mesenchymal stem cells in vitro.
