氰戊菊酯對(duì)小鼠精子獲能和頂體反應(yīng)的影響及其機(jī)制研究

【摘要】:第一部分 氰戊菊酯對(duì)小鼠精子獲能的影響及機(jī)制 一.小鼠精子獲能與鉀離子載體、鉀離子通道特性和作用 以金霉素(CTC)熒光染色法評(píng)價(jià)小鼠精子的體外獲能,并觀察鉀離子(K~+)載體纈氨霉素(Val)對(duì)精子獲能的影響。CTC熒光染色觀察,以B型精子發(fā)生率(%)評(píng)價(jià)獲能的程度,B型精子發(fā)生率越高,獲能越完全。Val試驗(yàn)以終濃度為50nmol·L~(-1)的Val處理精子,其B型精子發(fā)生率與對(duì)照組相比顯著增加(n=10,p<0.05),表明Val能促進(jìn)小鼠精子的獲能。由于Val主要具有促胞內(nèi)K~+離子外流作用,提示在精子獲能過(guò)程中,K~+離子是重要的一環(huán)。 進(jìn)一步采用全細(xì)胞膜片箝方法記錄小鼠精母細(xì)胞K~+離子通道電流,并分析其電流特性,同時(shí)觀察Val的影響。結(jié)果顯示:在小鼠粗線(xiàn)期精母細(xì)胞上能記錄到一個(gè)被緩慢激活的外向鉀電流,其幅度隨去
[Abstract]:Part one Effect of fenvalerate on sperm capacitation in mice and its mechanism I. Characteristics and effects of sperm capacitation and potassium ion carriers, potassium channels in mice The in vitro capacitation of mouse spermatozoa was evaluated by chlortetracycline (CTC) fluorescence staining, and the effect of valamomycin valeromycin (Vala), a potassium ion carrier, on sperm capacitation was observed. The higher the rate of capacitation, the more complete the capacitation. Val test was used to treat sperm with Val with the final concentration of 50nmol. The incidence of type B spermatozoa was significantly higher than that of the control group, indicating that Val could promote sperm capacitation in mice. Because Val mainly promotes intracellular K ~ + outflow, it is suggested that K ~ + is an important part of sperm capacitation. Furthermore, the K ~ + channel currents of mouse spermatocytes were recorded by whole-cell patch clamp method, and their current characteristics were analyzed, and the effects of Val were observed at the same time. The results showed that a slow-activated outward potassium current was recorded in mouse spermatocytes in the coarse line phase, and the amplitude of the current was removed.
